The paper focused on the karyotype analysis of five species maritime fish in the South China Sea, such as Nibea coibor, Trachinotus ovatus, Lutjanus sebae, Epinephelus tauvina and Rhabdosargussarba. Based on the above results the different bandings of Trachinotus ovatus were also researched. Karyotypes of five species fish were got successfully by analyzing the metaphase chromosome of them.These fishes all belong to Perciformes, and have become popular for its deliciousness, high value of nutrient and meat quality. The Nibea coibor' glue was valuable tonics of the traditional Chinese medicine. Lutjanus sebae was also used to see and enjoy. Rhabdosargussarba was raised not only in sea but also in cage. Trachinotus ovatus and Epinephelus tauvin were popular in many restaurants. In this paper, the karyotypes of them were reported for the first time. The results of this research work got some new information and provided convenience for the sea fish genetics and breeding.An improved new method of preparing chromosome specimen was obtained and used successfully, the experiment cycle was shortened. Kidney cells were separated from the living fish quickly then PHA and colchicines were added, After Giemsa staining and air drying, Karyotypes of five fishes and their karyotypes graph were completely gained. They are all diploid chromosome number.The karyotype formula of Nibea coibor was 48t, NF=48.Trachinotus ovatus was 8m+40t, NF=56.Lutjanus sebae was 2sm+46t, NF=50.Epinephelus tauvina was 48t, NF=48.Rhabdosargussarba was 4m+44t, NF=52.In addition, the Ag-NOR, C-bands, G-bands and R-bands of Trachinotus ovatus were got. The atlas showed that Ag-NORs always appear in the short arm of the middle centromere chromosome, while the frequency of 2 Ag-NORs were highest. Parts of the chromosome were C-band positive, and they were composed of centromeric, termination and interstitial. There was 1-3 G-bands on the chromosome arms. And R-bands were found in minority termination and interstitial of the chromosome.Based on the researching of the chromosome of the five sea fishes, not only the clear intermediate stages of chromosome were obtained, but also the karyotypes of them. Besides these, different kinds of chromosomes were discriminated exactly by initial analyzing the bandings of Trachinotus ovatus, which made karyotype analysis much more standard. These consequences provided convenience for the work of genetic breeding of Trachinotus ovatus, as well as offered useful informations for banding analysis of other sea fishes. |
