High Slope Pig Karyotypes And Banding Pattern

The material of this study was Gaopo Pig, and the compared group was Jianbai Xiang PigⅠline. This paper discusses the differences and commonnesses between the two kinds of pigs by using the technique of peripheral blood lymphocytes culture. Their fittest condition of culture for C-banding, G-banding and siver staining were found . This paper offered the basic materials for the exploitation,utilization and species of protection on Gaopo Pig. The results were as follows:1.The fittest conditions of culture by the method of peripheral blood lymphocytes culture for Gaopo Pig and Jianbai Xiang PigⅠline: The fittest PH of synthetical culture medium was 7.0～7.2, added Colchicin after cultured 68～68.5 hours, the final concentration of Colchicin was 0.04 ug/ml, then cultured 2～2.5 hours to Continue, 25～30min was took under the condition of KCl hypotonic treatment for the same time, then obtained favourable chromosome samples by fixing in fixed fluid and drying in air.2. The diploid chromosome number of Gaopo Pig and Jianbai Xiang PigⅠline were 2n=38, the same as other Susscrota domestica, the karyotypes of them were: 10sm+4st+12m+12t.3. The positions of C-banding and the character of banding patterns of Gaopo Pig and Jianbai Xiang PigⅠline were the same almost, the C-banding frequencies ocured on Gaopo Pig and Jianbai Xiang PigⅠline had some differencies. The frequencies of 3,5and 8 chromosomes ocured on Gaopo Pig were higher than Jianbai Xiang PigⅠline, it indicated that there were variabilities and commonnesses between Gaopo Pig and Jianbai Xiang PigⅠline, and it was used as the identify species. 4.This study obtained 54 regions of G-banding, 212 bandings in the middle age of Gaopo Pig, and 205 bandings of Jianbai Xiang PigⅠline. It demonstrated that the number of G-banding on Gaopo Pig was the same as Jianbai Xiang PigⅠline.5. The average number of Ag-NORs of Gaopo Pig (2.93) was lower than the number of Jianbai Xiang PigⅠline (3.97) ,The difference were significant (P<0.01).
