China is a mighty agriculture country. Wheats is Chinese main corps, and it's very important to the development of Chinese agriculture . Using heterosis of wheat have made our country's agriculture developed expeditiously . With the progress of agricultural technology , more and more wheat hybrid seeds are helpful to make ours agriculture high-product, stable product and high-quality . At the same time , the problem that identified the facticity and purity of wheats' hybrid seeds is deeply drew attention , and low-quality hybrid seeds would lose heterosis . Therefore , seeds' purity identification is an importance duty .In order to find a quick , practical and effective skill to indentify seeds purity of the new chemical hybrid wheat, cultivar XZ1 and XZ5 were studied through a few ways . The results were as follows :1. The way to identify seeds purity of the cultivar XZ1 and XZ5 through phenol dyeing is fast, but it is limited in the field of commerce circulation because of its high subjectivity;2. The way of seeds purity identification through polyacrylamide gel electrophoresis ( PAGE ) of EST isozyme in single seed is practical and effective to cultivar XZ5 , but it is not practical and effective to cultivar XZ1 , because it can not make a obvious difference between hybrid F1 seed and its maternal parent;3. The way of seeds purity identification through sod. dodecyl sulfate polyacrylamide gel electrophoresis ( SDS-PAGE ) of high-molecular-weight glutenin subunit ( HMW-GS ) in single seed can distinguish XZ1 and XZ5 hybrid F1 seeds from their parents , but it is also limited in the field of commerce circulation because of their unabundant polymorphism;4. The way of seeds purity identification through acid polyacrylamide gel electrophoresis ( A-PAGE ) of gliadin in seed is a quick , practical and effective method , and it can obvious distinguish XZ1 and XZ5 hybrid F1 seeds from their parents . To prove its feasibility , factitious intermixed seeds and chemical hybrid seeds were studied with acid polyacrylamide gel electrophoresis (A-PAGE) . The result shows that acid polyacrylamide gel electrophoresis (A-PAGE ) of gliadin in seeds' purity identification is very effective .5. Also, 43 cultivars of common wheat were studied with acid polyacrylamide gel electrophoresis (A-PAGE) . The differences of gliadin bands in 43 cultivars were significant. The' results indicated that acid polyacrylamide gel electrophoresis ( A-PAGE ) of gliadin is a quick , practical and effective method in seeds purity identification not only to the cultivar XZ1 and XZ5 but also to other common wheat cultivars. |
