This research regards ovule of Eucalyptus 12ABL as material,Studies on endosperm to regenerate plant in vitro culture of Eucalyptus 12ABL, relevant problems such as the best period that the endosperm in vitro culture,callus induction,inducing bud,inducing root,transplantation.1. Observe the processes of the early development of endosperm,The result of study shows :endosperm after pollinating 1~7d is the early development stage; endosperm after pollinating 8~12d begins to grow; endosperm after pollinating 13~16d grows more vigorous; endosperm after pollinating 17-20d grow slower; endosperm after pollinating 21~24d begins to degradate.2. Establishment of regeneration system from endosperm of Eucalyptus 12ABL: Studies have suggested drawing materials best period and moderate disinfecting method;It is best to deal with of materials for 3 days at low temperature;A modified medium consists of MS+BA1.5mg/L+IAA1.0mg/L+sugar 40g/L+agar7g/L that can get embryogenic callus;The optimal medium for inducing buds was MS + BA1.5mg/L + IAA0.1mg/L+sugar 40g/L + agar 7g/L;The optimal medium for multiplication was MS +BA1.0mg/L + NAA 0.2 mg/L+sugar 30g/L + agar 7g/L;The optimal rooting medium was 1/2MS +IBA0.5mg/Lsugar 30g/L + agar 7g/L.3. By comparing the triploid plants with normal liploid,leaves of triploid plants were thicker,larger,more green in color;and with larger stomata and less in number;the chromosome number of triploid plants was 2n= 33.
