Strawberry(Fragaria ananassa Duch.)belongs to the Rosaceae(Rosaccae)Strawberry is QFragaria),herbs,creeping plants.Origin from South America,is widely cultivated around the world.And it is the world's most important economic berries.Traditional breeding methods takes longer time and a lot of difficulties.The rise of molecular breeding provides a new solution for strawberry breeding.For Strawberry transgenic,the most commonly used method is Agrobacterium mediated transformation,Agrobacterium mediated transformation needs efficient strawberry callus culturing and re-differentiation system.Tissue culture pollution,slow callus growth,slow redistribution,long experimental period,has been important issues for the transgenic operation.It is of great significance to find the cause of pollution to further reduce the pollution problem,to screen the different varieties of strawberry cultivars with differentiated differentiation ability and to construct the efficient adventitious buds of strawberry and shorten the test cycle.The main findings are as follows:1.We screened two strawberry strains with high relative growth rate of NOR156,NOR164,NOR1,NOR 118,NOR 147,ES8,ES6,ES 12,LIT1,LIT7,FIN53,SL01,IT18,IT20,IT1 and IT6.2.This paper explores the causes of pollution in tissue culture.By observing the pollution of 397 medias and summarizing the reason of pollution,it is found that the ex vivo time and the health of the explants will affect the pollution rate and the disinfection efficiency to a great extent.Select the relatively robust explants,shorten the time in vitro,reduce the loss of explants,reduce the experimental operation and detonation process explants damage,reduce the number of wounds can effectively reduce the pollution rate.The concentration of disinfectant should also be related to the robustness of the explants,and test the effective disinfection method taken in this test:rinse 1h,flush with sterile water once,2%sodium hypochlorite solution disinfection 3min,sodium hypochlorite disinfection,Rinse with sterile water 3 times,40%of the PPM solution soak for 30s.3.The callus growth rate and the regenerative ability of adventitious buds were observed by the difference of the morphological differences between the tissue culture seedling leaves,the tissue culture seedlings petiole,the cuttings leaves,the cuttings petiole.It was found that the cuttings of the cuttings had a certain adventitious buds.NOR156,NOR164,NOR1,NOR 118,NOR 147,ES8,ES6,ES 12,LIT1,LIT7,FIN53,SL01,IT18,IT20,IT1 and IT6 of all 16 kinds of strawberries in the comparison of the difference of callus and adventitious buds.ES8 type of strawberries in the various types of explants from different sources,have a relatively prominent growth of adventitious buds.In the relationship between SOD and POD of strawberry seedlings,SOD was not significantly different.In the POD test,ES8 also showed a relatively high POD content.
