To determine the effect of cellulase on cellulose degradation in straw, series experiments were carried out to detect the submergered fermentation conditions and characteristics of enzyme both for Trichoderma viride 3.3711 & Trichoderma koningii 3.2774 in liquid state. The results showed that:(1) optimum conditions for enzyme yielding of Trichoderma viride 3.3711 were to use corn straw powder (demolition) as carbon source and the maximum of CMCase, filter paper andβ-glucosidase activity were 308.56U/mL,33.56 U/mL and 14.68U/mL respectively. ammonium sulfate as nitrogen source ,at pH value 5, with 5% of inoculation, at rotational speed of 150 r/ rain, temperature of 28℃, totally incubation for 144 h. The optimum enzyme active was under temperature between 50℃with pH of 4.5~5.0.Enzyme active in kept stable under 45℃,PH between 3.5~6.0. The optimum conditions for enzyme yielding of Trichoderma koningii 3.2774 were to use rice straw powder(blow up) and the maximum of CMCase, filter paper andβ-glucosidase activity were 276.36U/mL,37.88U/mL and 25.35U/mL respectively, the others were as same as for Trichoderma viride 3.3711.(2) A L16(45) orthogonal experiment design was used to detect the enzymatic degradation on straw, the factors were: primary enzymatic products, processing duration, raw materials and physical conditions. The results showed that the importance order for above tour factors was processing duration > raw materials> primary enzYmatic products >PH value. The best condition for straw degradation by cellulose was to add eight ml of Trichoderma viride 3.3711 into fine corn flour without any pH adjustment and incubate for 16 hours, and obtained relatively higher acidic fiber degradation rate.
