Resource and environmental issues have been the pivotal challenge of the development of modern society. The photosynthesis production of the earth amount to hundred millions tons, and ninety percent of which are cellulose. However, these recourses have not been fully exploited. Transforming cellulose into reproducible resources is one of the most important ways to solve world spread problems including shortage of food, environmental pollution, energy crisis, and so on. Research on molecular biology and genetic engineering of cellulose enzyme would definitely facilitate the process of cellulose application in industry and agriculture. It would transform cellulose that were wasted in the past into various materials that are useful in people's daily life using cellulose enzyme and improve the utilization of cellulose, which may solve crisis all over the world in the future.The cellulose research has great practical and application background for effective useing of cellulose, solving the cellulose pollution and waste, and suppling bioenergy resourse.Endoglucanase is the important components of the cellulase complex, then hydrolysis of 0-1, 4-glycosidic bond, the long-chain cellulose molecules truncated, resulting in a large number of non-reduction end with a small molecule of cellulose. It is energy efficient use of bio-cellulose occupy an important role. The ultimate goal of this study is to cut through the internal transformation of glucanase, to find the effective conversion of cellulose, and effective use of a large number of cellulose wasted resources, the degradation of cellulose can be used for human resources.In this paper, the specific work as follows:1. Using RT-PCR method, endo-glucanase gene (EGII) has been cloned from the Trichoderma viride. The endoglucanase gene (EGII) no intron, the sequence full-length is 1257bp,and amino acid 418.2. Through adding endoplasmic reticulum stranded signal in EGII gene, construction of reorganization vector of p3300-35S-EGâ…¡-Tnos, then transferred and resistanted plant regeneration to tobacco.3. Then transferred and resistanted plant regeneration to tobacco, and get the resistant plant of 33.
