The fermentation condition and coacting agent of Baclillus subtilis B2 was optimized and the colonization dynamics of that of around the rhizosphere of Angelica sinensis, Astragallus mongholicus and tomato, the disease prevention of the root rot of Angelica sinensis and Astragallus mongholicus were studied, The results were as follows:1 The optimization of fermentation condition of B2The two perfect culture medium, D-â… and D-â…¡were optimized, the D-â… was composited of glucose 1.0%, beef extract 0.8%, yeast extract paste 0.5% , pH 8.0. In the same culture condition, the active bacterial quantity of B2 in the D-â… differed significantly from that in the D and NB and was 2.05 and 52.36 times than that in the D and NB respectively ;The D-â…¡was composited of white granulated sugar 2.0%, beef extract 0.8%, yeast extract paste 0.35%, pH 8.5. There was no significant difference between the active bacterial quantity of B2 in the D-â…¡and D, while the cost of D-â…¡was cheaper than that of D. Meanwhile, the optimum match, 100ml inventory/250ml flask matching to the rotate speed of 140rpm, was screened and the the moderate fermention time 36 h was confirmed.2 The colonization dynamics of B2 around the rhizosphere of Angelica sinensis, Astragallus mongholicus and tomatoB2 could colonized strongly around and on the root of Angelica sinensis and Astragallus mongholicus, the active bacterial quantity where fell, rose and fell, the order of magnitude went down merely from 6 to 5; There was the similar colonying trendency of B2 beween around and on the root of Angelica sinensis and Astragallus mongholicus, while it could colonized faintly around and on the root of tomato, the active bacterial quantity where fell on the the root of tomato, the order of magnitude went down from 6 to 2.B2 could survive in the Angelica sinensis and Astragallus mongholicus plots designed for the colonization experiment from winter, the one order of magnitude merely fell from 4 to 3, but, the active bacterial quantity can not be detected after winter.3 The pesticide screened for the root rot of Angelica sinensisThe effect of fifteen-fold fermentation broth of B2 used for immersing seeding on the yield of Angelica sinensis was best and the yield was increased by30.7%, there was significant difference beween which and the other eight treatments, the one thoused times liquid of 1.8%AVM used for irrigating root and the the fifteen-fold fermentation broth of B2 used for irrigating root could increased the yield by 15.64% up to 18.48%; The prevention effect of one thoused times liquid of 1.8%AVM used for immersing seeding on the root rot of Angelica sinensis was best and by 79.05%,there was significant difference beween which and the other six treaments, the better treatments for disease control were fifteen-fold fermentation broth of B2 used for irrigating root and the one thoused liquid of 1.8%AWM used for irrigating root, whose control effect were 69.22% and 66.63%, respectively.4 The prevention experiment of the root rot of the Angelica sinensis and Astragallus mongholicusThe fifteen-fold fermentation broth of B2 used for immersing seeding matching to3kg/667m2 3% phoxim used for treating soil could increase yield of Angelica sinensis and Astragallus mongholicus by 35.07% and 37.25%, respectively, there was significantly different between which and the other three treatments;The prevention effect of nine hundreds times liquid of 20% Cupric acetate used for immersing seeding matching to 3kg/667m2 3% phoxim used for treating soil and3kg/667m2 3% phoxim used for treating soil on the root rot of the Angelica sinensis were 82.19% and 81.33%,respectively, they were not significantly different, but the prevention effect of which on the root rot of Astragallus mongholicus were 80.3% and 73.4%, they were significantly different.5 Study on the seed- coated agents composed of B2The promotion effect of 0.5% coated seed samples and 0.5% filme-coating agents on the germination of corn seed was obvious; 0.2% filme-coating agents reduced the germination vigor of melon seed, while it could not influence the gernimation rate of melon seed; 0.08% coated seed samples and filme-coating agents could not influence the gernimation of tomato.
