According to the tests of fermentation temperature, pH and ventilation size on Serratia plymuthica Z10,while its antagonistic mechanism,field trial of disease-control and seed coating agent were explored.The results are as follows:1 Studies on Fermentation Technical Parameters of Serratia plymuthica Z10The experiments in shake flask based discussion group about Z10 on marcescens univariate fermentation process parameters to explore,and experiment with the fermentation tank measured growth curve of Serratia plymuthica. The results indicated that the optimum incubated temperature, pH, and ventilation size were 28℃, 7.5 and 1000L/h, and added 10% culture media after 12 h fermented, and it showed that after 4 h and 16 h culture the bacteria arrived logarithmic phase and stabilization phase respectively, and the maximal concentrations of live bacteria was 10.632×1012 individuals/mL in 22h.Control of fermentatation period reasonable in the 22h.2 The experiments of antagonistic effection of Z10The experiment by face-to-face-culturing, select Rhizoctonia solani Kühn, Sclerotinia sclerntiorum (Lib.), Botrytis cinerea Pres.as pathogenic fungi .it showed that the Z10 has the inhibitory action to several kind of pathogenic fungi.and the inhibitory effect of fermentation fluid in 16h to 20h is the best time. The microscopic characteristics of antagonistic mechanism were showed: the hyphy twisted and deformed various structures,then the cells degenerated;the hyphy wall was destroyed and broken,the cytoplasm oozed out of the cell,the hyphy wall disintegrated and the cells collapsed.3 The experiments of field growth promotion and disease-control on Z10The experiment result of the field trial of disease-control of Solanum tuberosum showed, the production increase effect of Z10 handling of the potatoes the increased production rate and the control effect respectively 16.63% and 32.91% compared to ck, In addition,the experiment result on the yield of Angelica sinensis showed that the Z10 20 times of fluid processing both prevented disease well and increased production,the increase production rate and the control effect respectively 26.27%and 52.12%,and the proportion of the first class was the first.4 Study on the seed-coated agents composed of Z10With the single factor screening of 4 kinds of adhesive agent, 2 kinds of filme-coating agent, 3 kinds of suspending agent and 9 kinds of warning color,we can made sure that the seed-coated agents composed were adhesive agent Z-4, filme-coating agent C-1, suspending agent R-1 and warning color blue or purple,and its corresponding percentage is 2%,2%,0.5% and 0.5% respectively.With the plate-counter method,the quantity of live bacteria were measured after combinated the fermented broth and coated seeds with different proportions in 1d,10d,30d,60d and 90d respectively.The result showed that the quantity of live bacteria of Serratia plymuthica Z10 and Bacillus subtilis B2 on the surface of coated seeds declined within 10d,then 10d later tend to stable,that is, over 105cfu/ml;When arrived at 90d, on the surface of coated seeds of Z10 fermented broth and coated seeds with the proportions 7:50 the quantity of live bacteria was highest(5.87×105cfu/ml ); the proportion 1:10 of B2 was highest(6.21×105cfu/ml ).Therefore, to applicate the seed coating, the best preparation is to additived seed-coated agents before, and then sowing proportion within 10d after adding bacteria coating, this is good for coating more quantity of live bacteria, improving the effect of growth promotion and disease control on seed-coated agents,storing for some time after coated as well.5 Effects on plant germination of Z10In three kind of plants′seed-coating germination test,the rate of Z10 and B2 coated seed of corn is 63% and 58% and the Ck is 61%; the rate of Z10 and B2 coated seed of tomato is 83% and 85% and the Ck is 86%; while the uncoated rape seed-germination percentage has not achieved 100%, Z10 and the B2 coating seed-germination percentage has also achieved 100%. Therefore, biological seed coating Z10 and B2 had no effect on seed germination and can be widely applied in production.
