In this study, different effects of PSⅡinhibiting herbicide atrazine were studied in Triticum aestivum and Chinese cabbage through physiology detection, 2D/SDS-PAGE and mass spectrometry.Effects of atrazine on physiological indexes were detected in Triticum aestivum after treatment with atrazine at different concentrations for 5 and 10 days respectively, and in Chinese cabbage for 4 days. The results indicated that the pigment content and soluble protein increased when the concentration of atrazine was below 10mg/L; when the concentration of atrazine beyond 10mg/L, the pigment content of both crops were significantly decreased. So 10mg/L was believed to be the threshold concentration of atrazine for these two crops.2D/SDS-PAGE analysis found that the composition and contents of total protein were changed evidently after treated with threshold concentration of atrazine in Triticum aestivum and Chinese cabbage. 3 protein species were lost, 5 protein species were newly induced in Triticum aestivum; 2 protein species were lost, 1 protein species was newly induced in Chinese cabbage. These protein species were all at low and middling molecular and had wide range at pI.2D/SDS-PAGE analysis indicated that the composition and contents of chloroplast proteome were changed significantly after treated with threshold concentration of atrazine in Triticum aestivum and Chinese cabbage. After treatment with 10mg/L atrazine, the protein species of two crops were lost or decreased. When the concentration of atrazine was under threshold, the composition and contents of chloroplast protein species of Chinese cabbage were increased. MS analysis on these changed protein species indicated that they were enzymes, which played important roles in photosynthesis. They were 2 activase isoforms of RuBPcase,Rubisco activase beta form precursor,23 kDa oxygen evolving protein of photosystemⅡ,3—phosphoglycerate kinase,Carbonic anhydrase, chloroplast precursor,glycosyl transferase,Isoprene synthase,proline dehydrogenase,6-phospho-fructo kinase isozymeⅡ,calcium transporting ATPase,maturase K.
