The Effects Of ELP,F-188 And CMG,LMS,on The Emulsification And Immunization Of FMDV Vaccine

Objective: The purpose of this research is to study the effects of emulsifier ELP,F-188 on stability of FMDV vaccine emulsion and the effects of immunity intensifier CMG,LMS on immunization of O FMD (HK)vaccine. The emulsified craft and the non- specificity immunity enhancement factor of FMD vaccine were studied in order to improve the emulsified craft and enhance the immunity potency of vaccine.Methed: The orthogonal experiment tables were constructed L9(34), the emulsifier F-188, ELP and Tween-80 as the factors. The 1-9# mixesemul sifiers formula was obtained. Refering to mutually emulsified method and pre-test result, mix emulsifier,white oil and the DMEM according to the proportion. It was emulsified for 5min in advance, then emulsified it for 20min in the high-speed mulser. A sample was taken, and the physical character (Emulsion outward appearance,dosage-form,stability,viscosity,emulsion particle size and so on), were valued. The most outstanding mixing emulsifier formula was chosen.The O foot-and-mouth disease virus were raised by BHK-21 cells conventionally, and were multiplied. Virus poisonous price was determined through micro method. Inactivate the venom whose log10TCID50 was higher than 7.0, with 0.2 mol/L BEI for 24-28h in 30℃. After examined in the young mouse and determined safe, matching vaccine was carried.The immunity intensifier CMG,LMS were separately established in three monitoring teams by 0, 100, 200 mg/kg body weight and 0,20, 40 mg/kg body weight (in Balb/c mouse's). The immunity intensifiers were added into virus-liquid according to the proportion and vaccine was preparared. After the safe examination, Balb/c mice were inoculated(0.2ml/one mouse). Intensive immunity on the 28th day, tail of mice was cutted and blood was picked at 14th, 21th and 28th day after the first immunity and 7th, 14th, 21thday after the intensive immunity and blood serum was separated. The level of anti-body was measured with LPB-ELISA. On 21th day after the intensive immunity, the phagocytosis of mononuclear phagocyte system was determined by Carbon particle clearance test.Result: The results showed that the different dosage three kind of emulsifiers (F-188, ELP, Tween-80)have the extremely remarkable influence the stability of the vaccine emulsion. The best of proportion was 1:3:3. In addition, optimal dose of Tween-80 was joined into the oil phase could enhance emulsified effect of emulsion; they were emulsified and vaccine was prepared. Inactivated venom of O FMDV was mixed with the different dosage immunity CMG,LMS. Vaccine immunity function was determined by experimental animal Balb/c mice. The results showed that CMG,LMS could non-specificity enhance experimental animal's immunity .But the separate effect was better than united one.
