It is possible to ectogenesticly culture the cell, tissue, apparatus of the plant or the plant with the developing of the biology technology. The cell culture has wide spread prospect. The plant cell has the almightiness of biosynthesis. Every cell retains a suit of complete genetic information. So the plant cell maybe produces the plant's compound. At present, the mulberry was studied on the quick reproduce, the undifferentiated tissue developed, and the new mulberry breed by the gene, and so on. It is not reported about the mulberry cell suspension culture.We researched on the establishment of mulberry suspension cell system, and its physiological characteristic; the change regularity of pH and central element in the culture medium; the growth effect of mulberry suspension cell by the different phytohormones .The main research findings are as follows.1. The mulberry suspension cell systems of Da10, Shinichinose, ShengDong No.1, were cultured; we studied on the growth rule and the physiological characteristic of mulberry suspension cell. The tissue culture plant of Da10, XiYiZhiLai, and ShengDong No.1 was used to develop the undifferentiated tissue. The 3 kinds of mulberry undifferentiated tissue were used to found mulberry suspension cell system after 4 cultured time in the B5, N6 and MS liquid mediums. The growth regularity is 14 d. It has 4 growths stages: the lag phase, the linear phase, the slow phase, the retardation phase. The B5 liquid medium was suited to the mulberry suspension cell. The mitotic index of mulberry suspension cell is lower in the lag phase than other phases. The most powerful viability happened during the lag phase. The inoculation quantity of mulberry suspension cell is 3 g·L-1 - 12 g·L-1 in the B5 liquid medium.2. The change regularity of pH and central element in the culture medium was researched. The result indicated that: the pH drops quickly in the lag phase, gradually rises afterwards, finally stabilizes about 5.38 as a result of the suspension cell physiological function; The suspension cell only consumes a little sugar in the lag phase, and the content of sugar drops in the linear phase; The suspension cell makes faster use of the NH4+ than NO3-; The mulberry suspension cell consumes the PO43- in the culture prophase.3. The growth effect of mulberry suspension cell by the different phytohormone was studied. The different phytohormone has different influence in the mulberry suspension cell culture process. The growth speed of mulberry suspension cell single using NAA 0.5 mg·L-1 is quickest than single using other phytohormones. The growth speed of mulberry suspension cell using 6-BA 1.2 mg·L-1 + NAA 0.1 mg·L-1 is quickest than other the combination of phytohormone. But different kind of mulberry should use different type and concentration of phytohormone. The combination of 6-BA 1.2 mg·L-1 + 2, 4-D 0.2 mg·L-1 was suited to the Da 10, and the combination of 6-BA 1.2 mg·L-1 + 2, 4-D 0.1 mg·L-1 was suited to the Shinichinose, and ShengDong No.1 in the combination of 6-BA and 2, 4-D. The result shows that more types or concentration of phytohormone was impossibility suited to the mulberry suspension cell. The different phytohormone should be used to the different kinds of plant.
