The peony is one of world famous flower and is widely loved in china,with a long history of cultivation and deep cultural background. For a long time, the improvement of varieties and breeding work has been the main content of peony research and production. Peony are good material for polyploid breeding with lower chromosome multiple and less number. polyploid breeding can improve Peony's watches quality and economic value.The experiment uses colchicin as main inductive medicament and Dimethyl Sulphoxide as penetrant, through soaking seeds, packaging, absorption and embrionic induction and soon the method to induce seeds,buds and embryos of Paeonia ostii 'Feng Dan', Paeonia suffruticosa 'Wu Long Peng Sheng'and Paeonia rockii hybrids. The auther expected to find out a suitable inductive method to have the polyploid and to lay certain foundation for peony's polyploid breeding.The results of this experiment as follows:1. Through soaking seeds we obtained 60 sprouts of P. ostii 'Feng Dan' and 37 sprouts of P. rockii hybrids in 2006, and 86 sprouts of P. ostii 'Feng Dan' and 124 sprouts of P. rockii hybrids in 2007. Carrying the morphology observation and variance analysis using the Duncan method on the seedlings of 2006, We had not discovered the obvious difference. The blade width of P. ostii 'Feng Dan' had certain individual difference and formed 3 ranks, but they had not obvious difference with comparison.2. Through packaging to separate buds of P. ostii 'Feng Dan', P. suffruticosa 'Wu Long Peng Sheng'and P. rockii hybrids we obtained 83 survival plants of all the peonies, and 54 of all the peonies by absorption method. At initial period of sprout, carriying on the cytology appraisal to the young leaf apex, we had not discovered the polyploid. The peony had not blossomed in the entire growth process and had not appeared the variation phenomenon.3. Through packaging to buds of P. ostii 'Feng Dan', we obtained 247 survival plants of all. The buds displayed normal, demaged and died three kind of growth conditions. The processing combination caused the best inductive effect was: the colchicin density was 0.2%, the Dimethyl Sulphoxide density was 1%, the processtimewas 3 day-long processing combinations, inductive rate achieved above 30%. Carrying the morphology observation and variance analysis using the Duncan method on the leaves, we discovered that: the blade width obviously enlarged, but the blade longth had the tendency which shortened slightly.4. Through culture medium plunging to the embryos of P. ostii 'Feng Dan' we had not achieved the ideal effect,because in the growth process the embryos successively all appeared the turning-brown and the death phenomenon. Through packaging to the embryos of P. ostii 'Feng Dan', we obtained 61 inductive plants. The processing combination caused the best inductive effect was: the colchicin density was 0.5%, the Dimethyl Sulphoxide density was 1%, the processtimewas 3 day-long processing combinations, inductive rate achieved above 20%.
