Flesh color is a very important trait related to the quality of nectarine fruit, And it can be used in the study of assisted breeding of nectarine. With the population of 89 hybrid plants derived from the intraspecific cross between nectarine variety"qinguang"and"shuguang", and by RAPD technique and BSA , The trait of flesh color of nectarine was studied.The main fruits of this research were:1,The method of extracting DNA using CTAB was improved. In this research ,the time of low temperature treatment for nectarine materials in liquid nitrogen was prolonged; and though the extracting buffer was not added PVP and 2-Mereaptoethanol,purified with phenol/chloroform,we gainned the high quality genomic DNA. It revealed that the effect of polyphenolics and polysaccharides to the quality of DNA could be reduced by prolonging the time of low temperature treatment.2,Two molecular markers linked to the gene of flesh color trait of nectarine fruit, s21-400 and s486-2000, were obtained. In this research,the molecular markers linked to the gene of flesh color trait of nectarine fruit were detected by the molecular biotechnology of RAPD and BSA method With the population of 89 hybrid plants derived from the intraspecific cross between nectarine variety"qinguang"and"shuguang".Two molecular markers linked to flesh color of nectarine fruits were revealed from 340 RAPD primers. By making a genetic linkage map of flesh color using the two markers and other markers reported, we found the best RAPD marker s21-400, it's genetic linkage distance is 14 cM.3,The effects of double primers RAPD was analyzed. Five combined primers were used to the amplification of nectarine cultivar'Shuguang'. The results showed that: there was no new stripe appeared in these five combined primers, and the results of the amplification was simply the addition of single primer.
