Utilizing one step unilateralism continuous sodium dodecyl sulphate polyacrylamide get electrophoresis (SDS-PAGE) technology study different effect facts in separate High-Molecular-Weight Glutenin Subunits (HMW-GS) and Low-Molecular-Weight Glutenin Subunits (LM W-GS) of wheat and improving the technology. Then using this way to analysis the hight and low glutenin subnits of breeding material of shaanxi province and the inheritance of F1 of crossbreed wheat ,the result show as flow :1. It can have a satisfactory SDS-PAGE result of HMW-GS and LMW-GS by the way with a appropriate extraction method and in the condition of 4.8% concentrated colloid, 12.0%-14.0% continuous gradate separate colloid, 35-40mA electric current and electrophoresis 330min. With the way it can avoid the effect of albumin and globulin to glutenin subunit separate. At the same time ,it can analysis many samples in a separate colloid.2.There are ten kinds allelic genes loci in Glu-1 and have twelve kinds combination form. Null and 1 loci in Glu-Al, the frequency are 22.5 percent and 77.5 percent. In Glu-Bl there have six kinds allelic genes. They are 7,7+8,7+9,14+15,13+16 and 17+18. 7+8 have the highest frequency which is 47.5 percen , and 13+16 have the lowest frequency which is 2.5 percent. In Glu-Dl there have tow kinds allelic genes. They are 2+12 and 5+10. The frequency is 72.5 percent and 22.5 percent.3. There are eleven kinds allelic genes loci in Glu-3 and have nineteen kinds combination form. Four kinds loci in Glu-A3. they are Glu-A3a,Glu-A3b,Glu-A3c and Glu-A3d. And the frequency are 40.0 percent, 12.5 percent, 10 percent, 37.5 percent. In Glu-B3 there have seven kinds allelic genes. They are Glu-B3a,Glu-B3b,Glu-B3c,Glu-B3d,Glu-B3f,Glu-B3h and Glu-B3j. Glu-B3d have the highest frequency which is 40.0 percen , and Glu-B3c and Glu-B3h have the lowest frequency which is 5.0 percent. In the nineteen kinds combination form Glu-A3a+Glu-B3d have the highest frequency which is 22.5 percent and there are nine material have the lowest same frequency which are 2.5 percent. There five variety haveGlu-A3d+Glu-B3d form which is a good quality subunit combination form.4.The result of Nei's genetic variation index show, at Glu-Al loci was 0.3488, at Glu-Bl loci was 0.6581, at Glu-Dl loci was 0.3750 , the average was 0.44606. And at Glu-A3 loci was 0.6738, at Glu-B3 loci was 0.8088.5. The inheritance analysis result showed that the band patterns of HMW-GS and LMW-GS were genetically controlled. The HMW-GS and LME-GS of wheat can be transmintted to the hybrid Fl as dominance from both parents and the phenomenon that HMW-GS and LMW-GS could not express was observed in F] of hybrids.
