Strains are one of the important means of production in mushroom industry, and its quality directly reflects the development of mushroom industry. In this thesis, tow Flammulina velutipes strains including F0005 and F0030 and two Pleurotus eryngii strains including Ple0002 and Ple0066 are studied from strains production,strains purity, and strains activity, which are used to establish complete strains quality control technology. Main results were as follows:1. Strains qualitystrains quality related to both nutrition conditions and cultivating conditions. The effects of different media carbon sources , water content, pH and temperature on mycelium growth , and fruitbody yield of Pleurotus eryngii and Flammulina velutipes strains were studied in this paper. The results showed that the effects of various media on mycelium growth rate and fruitbody yield were different . the formula with branches as the main material is the best for stock culture medium, the mycelium growth rate was faster ,fruit- body yield was one of the greatest the formula with cottonseedhulls as the main material on which the mycelium grew slower but growth potential of mycelium is stronger and fruit-body yield was the another greatest and the proportioning of cottonseedhulls need to be regulated in futher. I t was on kernel stock culture medium that the growth speed of mycelium is the fastest, the growth potential of mycelium is the strongest,but fruit- body yield was lower.strains quality was poorer ,which mycelium was cultured on the medium with sawdust, juncao or bagasse as the main material.For Pleurotus eryngii strains, the optimum water content of stock culture medium was 63%.The optimum pH was between 6.2-6.5 .The optimum temperature was 25℃. For Flammulina velutipes strains, the optimum water content of stock culture medium was 60%. The optimum temperature was 20-25℃. The optimum pH was between 6.2-6.5 . There are no significance difference about strains quality of stock culture medium between pH 5.1-6.75. But there are somewhat difference between strains F0005 and F0030.The results also showed that an appropriate temperature stimulation can improve mycelium adaptability to environment. All of stock culture within 110 days can be applied to production. There are no significance difference on yield.2.Strains PurityThe normal Flammulina velutipes strains without contamination were inoculated with different concentrations of E.coli in order to acquire strains with invisible bacteria pollution. Four kinds of methods were carried on the invisible contamination-strains to detect E.coli.The results were as follows:Four kinds of methods all could detect the presence of bacteria. Plate coating and liquid shake culture method had the highest sensitivity,it can detect the limit of 100 bacterias per- gram mycelium; streak plate method and the boiling PCR method could detect 10000 bacterias per -gram mycelium.Two Pleurotus eryngii strains of mycelium are used as material to tested from dsRNA viruses , dsRNA viruses is detected in the Pleurotusostreatus strains by means of dsRNA technology and validation while not detected in other two Pleurotus eryngii strains, which shows that this method can be used for edible virus detection.3. Strains ActivityStrains at different Preservation time are used as material to measure several physiological and biochemical indicators maybe related to strain aging .With results showing that:There are differences in appearance shape,but no significance difference in micro morphology and germinating colony characteristics for strains of different Preservation time.Among the activity of eleven enzymes. The activity of amylase , CMC enzyme , xylanase and acidic phosphatase have significant negative correlation with Pleurotus eryngii strains Preservation time. In which, the activity of laccase and polyphenol oxidase have significant positive correlation with Pleurotus eryngii strains Preservation time.For Flammulina velutipes strains , the activity of CMC enzyme, xylanase, laccase, the malondialdehyde and the pH of stock culture medium have significant positivecorrelation with strains Preservation time.And amylase,superoxide dismutase, peroxidase, hydrogenperoxidase have significant negative correlation with strains Preservation time. BTB can intuitivey indicate the changes of pH.In the high temperature resistance experiment, Along with the increase of Preservation time, high temperature resistance of strains decreased. In the antichemical medicines (ACR and ACT) of Pleurotus eryngii strains experiment. Along with the increase of Preservation time,the resistance of mother culture obviously decrease.Although the resistance of stock culture is stronger, the difference can detected by weakened growth speed.
