Armillaria(Armillaria spp.) is Eukaryota,Basidiomycota,Basidiomycetae,Hymenomycetes, Tricholomataceae,Armillaria (Fr.:Fr.) Staude.It was also known as Hazel mushroom Armillaria was a kind of edible and medicinal fungi.It contained a lot of chemical components. The chief chemical component of it was the polysaccharides. Over the years, the number of experimental studied had shown that Armillaria polysaccharides had a variety of biological activities.And many kinds of Armillaria preparations had been produced,but mostly belonged to the second generation of health care products.It was not exactly well studied that what was the effective ingredients of Armillaria's various function and where was the main active site.In this thesis,it reacearched that polysaccharide was purified from Armillaria fruiting bodies of artificial cultivation and antxidioant activities of polysaccharide.It was believed that it provided a theoretic basis in pharmacological effects of Armillaria polysaccharide and it had significant value in the prospect and development status of Armillaria.The main contents and conclusions in the research were as follows:1.Three extraction technology applied in the extraction of Armillaria's polysaccharide.With Armillaria powder as material,three different methods which contained hot water extraction,ultrasonic extraction and enzymatic extraction were investigated by orthogonal test method and optimized the extraction process.The results showed that the extraction of Armillaria'polysaccharide was 5.96% by hot water extraction,5.38% by ultrasonic extraction method and 6.25% by enzymatic extraction.And hot water extraction, ultrasonic extraction had characteristics of short extracting time,low energy consumption,higher efficiency,but enzyme extraction method can significantly improve the yield of polysaccharides.Therefore,the enzymatic extraction of polysaccharides was an ideal extraction method.2.Polysaccharides of Armillaria's fruiting bodies were purified. polysaccharide was graded to three components,T30,T60and T80, by alcohol precipitation. Two pure polysaccharides contained T301 and T302 was obtained by T30 gel column chromatography(sephadex G-150). The pure polysaccharides T601 was obtained by T60gel column chromatography(sephadex G-150).3.The antxidioant activity of Armillaria polysaccharides had been studied.the.T301 and T601 had been done experimental study on vitro antxidioant,and the result showed that two kinds of polysaccharides had certain antioxidant activity,that the effect of T301's was better than T601'.The biological activities of polysaccharide had certain relationship with molecular weight.4.Design formula.Recipe seasoning of Armillaria instant granules had been determined by sensory evaluation.Optimal formula was as follows:19.86%Armillaria puree,79.44%sugar,0.70%citric acid.
