Animal infectious diseases have always been one of the important factors restricting the development of animal husbandry in our country.In the process of disease prevention and control,disinfection as one of the measures to prevent the spread of animal diseases has attracted more and more attention.In this paper,through the research of compound disinfectants with quaternary ammonium salt as the main ingredient,the disinfectant with good sterilization effect and high efficiency is developed to ensure the sustainable and healthy development of the aquaculture industry in my country.In this study,eight single and double chain quaternary ammonium salts were used as raw materials,E.coli 8099,Staphylococcus aureus ATCC 6538,and Candida albicans ATCC 10231 were used as test strains.A good single and double chain quaternary ammonium salt is the pharmaceutical composition of the compound disinfectant.The single and double chain quaternary ammonium salt was tested by the chessboard method,and the best ratio was screened.The antibacterial test was used to screen the different p H and different chelating agents of the compound disinfectant to obtain the optimal p H and optimal chelating mixture.A compound disinfectant formula with a ratio of benzalkonium chloride and decamethonium chloride of 2 ∶1 ~ 1 ∶2 was successfully developed,supplemented by a new type of decamethonium chloride disinfectant with a p H of 9 and a chelating agent of 0.001% EDTA·2Na.In the evaluation of antibacterial effect,antibacterial mainly includes the preliminary selection test of neutralizer,the identification test of neutralizer,the qualitative sterilization experiment of suspension,the quantitative sterilization experiment of suspension,the comparison experiment of disinfection effect of new decylmethylammonium chloride disinfectant and other disinfectants,Experiment on the disinfection effect of multiple ingredients in the new decylmethylammonium chloride disinfectant.Finally,the neutralizer of the compound disinfectant was selected as a PBS solution of 0.5% lecithin + 3% Tween-80.E.coli 8099 and Staphylococcus aureus ATCC 6538 were used as the test strains.The suspension qualitative sterilization test was used to determine the lowest sterilization.Concentration and effective sterilization time,use quantitative sterilization test to determine the sterilization effect of compound disinfectant at a certain concentration.The results showed that the minimum effective bactericidal concentration of compound didecyl dimethyl ammonium chloride solution against Escherichia coli and Staphylococcus aureus were 1∶12 800 and 1∶25 600,respectively,and the fastest effective time for sterilization at this concentration was 10 min.The compound didecyl dimethyl ammonium chloride solution acts on Escherichia coli and Staphylococcus aureus for 2.5 min,5 min,and 7.5 min.The sterilization rate is 100%,and the killing logarithm is ≥ 5.0.Comparing the disinfectant effect of this compound disinfectant and the two listed disinfectants based on the concentration of quaternary ammonium salt,the disinfectant effect of compound decamethonium chloride and decamethonium bromide solution are both against Escherichia coli after being diluted with quaternary ammonium salt concentration as the benchmark.The sterilization effect of all is 99.99%,which is better than that of quaternary ammonium salt glutaraldehyde solution.When the three disinfectants acted for more than 2.5 minutes,the sterilization rate of Staphylococcus aureus was greater than 99%.Finally,the compound disinfectant and its decamethylammonium chloride and benzalkonium chloride are evaluated for the disinfection effect within a specific time,diluted with the quaternary ammonium salt concentration as the benchmark,and the sterilization effect on E.coli can reach100% after an action of 4.5 min or more.The bactericidal effect on Staphylococcus aureus can reach 100%after acting for 3 min or more.In summary,this study screened out a better prescription for compound decamethonium chloride disinfectant,and the compound decamethonium chloride disinfectant developed has a good bactericidal effect.
