| Schisandra as a member of Magnoliaceae are dried mature fruits.Because of its sweet and sour pulp, seeds ups, slightly salty, named for the Schisandra. It was sorted of Schisandra chinensen and kadsura longepedunculat. Schisandra especcially was Schisandra chinensen. Schisandra chinensen is precious chinese medicine with cure of the lung,nourishing the kidneys and calm the nerves.Pharmacological studies have shown that Schisandra can regulate biological systems of life,for example central nervous system, cardiovascular system,immune system and so on.Also, the anti-tumor, anti-oxidation, anti-aging, protect the liver are its effect.The main effective ingredients of Schisandra chinensen included lignans, polysaccharide and essential oil.In current,researhs of lignans were more and deepen study. Study of essential oil were focused on extraction and component analysis.Howerevr, biological activity and mechanisms of HepG2 apoptosis induced by essential oil have't been reported. And this is the purpose of this study.1 Studies on extraction and components identification of essential oil from Schisandra chinensenEssential oi was extravted by distillation-extraction and Compositions were analyzed by GC-MS and the major components are Ylangene(19.83%), Spiro[5.5]unden-2-ene,3,7,7-trimethyl-11-methylene-,(-)-(5.24%),Bicyclo[3.1.1]hept-2-e ne,2,6-dimethyl-6-(4-methyl-3-pentenyl)-(7.18%),1H-benzocyclohtene,2,4a,5,6,7,8-hexa hyydro-3,5,5,9-tetramthyl-,(r)-(5.71%),Benzene,1-methyl-4-(1,2,2-trimethlcyclopentyl)-, (r)-(5.58%).2 Studies bioligical activity of essential oil from Schisandra chinensenAntioxidant activity was measured using DPPH and ABTS radicals. Results shown the essential oil can reduce both DPPH and ABTS radicals formation in a dose-dependent manner. Moreover, the ABTS radicals scavenging is stronger than that of DPPH at the same concentration of Essential oil. EC50 of Essential Oil The essential oil exhibited higher EC50 value against DPPH radicals (1.49±0.08μL/mL) than ABTS (0.57±0.09μL/mL).Antimicrobial activity on Escherichia coli, Bacillus subtilis and Staphylococcus aureus was assay by filter paper measurement. The results shown that three kinds of bacteria were inhibited by essential oil with bacteriostatic circle diameter of 17.67±0.56,16.67±0.31 and 8.33±0.11mm.However, for E.coli and bacillus subtilis bacteriostatic effect significantly greater than staphylococcus aureusThe cytotoxicity of the essential oil on HepG2,SY5Y,B16 and HeLa was assayed by MTT,and with IC50 are respectively SY5Y (0.29±0.08), HeLa (0.20±0.05), and B16 (0.13±0.08).3 Study on HepG2 apoptosis indcued by essential oilThe essential oil from Schisandra chinensen decreased the cell viability in HepG2 human hepatoma cells in a concentration and time-related manner. In addition, The essential oil incresed production of ROS and induced HepG2 cells apoptosis.However, the essential oil reduced the activity of T-SOD.These results suggest essential oil made apoptosis by high level ROS or inhibit antioxidant defenses or their combinations.Therefore, the essential oil from Schisandra chinensen is a good candidate for hepatocellular carcinoma (HCC) therapy and that reactive oxygen species are the key signaling molecules in the essential oil-induced apoptosis in HepG2 cells. |
