Biological enrichment was adopted in this study. To choose barley seedling as an enriching selenium (Se) object and use its bioconversion which can transform inorganic Se to organic Se, it could achieve the double effect of barley seedling and organic Se. After establishing animal models, it could be observed of immune regulation function, hepatoprotective function, anti-ulcer effect, anti-tumor effect, toxicological effect of selenium-enriched barley seedling (SEBS) in mice.Immune regulation function of SEBS in mice: Experimental mice were orally administered with SEBS at doses of 0.1g/ml, 0.5g/ml and 1g/ml. Thirteen days later, total number of white blood cell (WBC), phagocytosis test of macrophage on abdominal cavity, titer of antibody, number of antibody-producing cells and weight of immune organs in mice were determined. The results suggest that compared to saline water group, SEBS could significantly improve phagocytic function of macrophages (P<0.01), obviously increase the titer of anti-CRBC antibody (P<0.05), spleen index (P<0.05), and total number of WBC (P<0.05), SEBS can promote cell-mediated immunity and humoral immunity in mice in a dose-dependent manner.. Hepatoprotective function of SEBS in mice with acute hepatic injury: The mice with acute hepatic injury induced by CCl4 were orally administered SEBS at different doses (0.1, 0.5, 1g/ml). Eighteen days later, the immune organs index and the activities of serum alanine aminotransferase (ALT) were investigated. On the other hand, the pathological changes of the liver were determined in all groups. The results suggest that SEBS at three doses significantly inhibited ALT activity in serum (P<0.01) and promoted the recovery of hepatic tissue injury, which was in a dose-dependent mode. Moreover, SEBS also obviously improved spleen index (P<0.05). SEBS has hepatoprotective function for mice with acute hepatic injury induced by CCl4.Effect of SEBS in mice with alcoholic gastric ulcer: The mice with alcoholic gastric ulcer were orally administered SEBS at different doses (0.1, 0.5, 1g/ml). The area of the ulcer was quantified by the methods of picture integration, weighing, marking and grading. Then the ratio of ulcer area to total stomach area (ulcer area ratio, UAR) was calculated to assess the degrees of the ulcers on the stomach wall. The results suggest that SEBS at three doses could significantly cut down the gastric ulcer (P<0.05), SEBS could reduce the alcoholic gastric ulcer in mice in a dose-dependent manner.Anti-tumor effect of SEBS in vivo and in vitro: The S-180 model mice were orally administered SEBS at different doses (0.1, 0.5, 1g/ml). Twelve days later, tumor weight and the immune organs index in mice were determined. Meanwhile, the inhibition effect of SEBS on the of mouse sarcoma cell S-180 and myeloma cell SP-2 were investigated in vitro by MTT assay. The results suggest that SEBS could significantly inhibit the growth of S-180 sarcoma in mice (P<0.05), obviously increase the spleen index and thymus index (P<0.05), obviously inhibit the growth of S-180 and SP-2 in vitro (P<0.05), which was in a dose-dependent mode. SEBS can suppress the growth of tumor in vivo and in vitro.Toxicological effect of SEBS in mice: Experimental mice were orally administered with SEBS at different doses (0.1, 0.5, 1g/ml) for twenty days, the acute oral toxicity test was conducted to understand the acute toxic effect, and the mouse bone marrow micronucleus and chromosomal aberrations tests were also conducted to investigate the genetic toxic effect. The results suggest that SEBS was safe with no acute and genetic toxic effect in the tested dose range.
