| Sorghum bicolor×S. Sudanense is an annual gramineous forage grass that mainly uses its stalks and leaves; it is the hybrid of sorghum and sudanense. Sorghum bicolor×S. Sudanense conbines the advantages of two parents, showing evidently intervarietal heterosis,and have expansive foreground in resource utilization and entironment protection.It combines advantages of the sorghum diameter,leaf width and Sudanense tillers, renewable and strong regeneration, heterosis is obvious. With drought resistance, heat resistance and more Cold-resistant, it have availability features which in precipitation or irrigation areas could get high yields and so on,are deeply welcomed by farmers.It has a broad application prospects in animal husbandry,aquaculture,and resource utilization and environmental protection.On the basis of the published sorghum sequence database, the SSR of Sorghum bicolor×S. Sudanense genetic markers was designed and their polymorphism was analyzed and evaluated in this study. The purpose of this study was to enrich the Sorghum bicolor×S. Sudanense's type and the number of molecular markers, to be the foundation for further development of the Sorghum bicolor×S. Sudanense genetic mapping and marker-assisted breeding, and to create the prerequisite conditions for further Sorghum bicolor×S. Sudanense important agronomic traits and QTL positioning.On the basis of gene bank of Sorghum, non-coding sequences were used as template for designing primers; according to e-PCR, the designed primers was validated and whether non-coding region of SSR loci's polymorphisms is higher than the coding region's polymorphisms was evaluated. In addition, the genetic diversity of 19 Sorghum bicolor×S. Sudanense varieties and 3 relatives of plant material were studied, according to the technology of SSR markers and these species and the genetic relationship among all the genetic varieties were discussed; the theoretical foundation was provided for the genetic breeding of Sorghum bicolor×S. Sudanense and further research.The experiment results are as follows:1. According to the e-PCR's analysis in sorghum, the frequency of dinucleotide repeats was the highest, the trinucleotide repeats was the second, accounting for 43.36% and 32.38% of the total SSR. By making a comparison of ten chromosomes in sorghum, the frequency of AT / TA, AG / TC, AC / TG, CG / GC, CT / GA, CA / GT, CGC / GCG, CGT / GCA, CGA / GCT, GGA / CCT were higher.To select 50 pairs of primers were synthesed. And take the Sorghum bicolor×S. Sudanense varieties like Xian-Feng and Le-Shi as template, of which 8 pairs of primers showed polymorphism, accounting for 16%.2. Through 86 pairs of polymorphic SSR markers to calculate the 19 Sorghum bicolor×S. Sudanense varieties and 3 relatives of plant materials,the genetic distances ranging from 0.42 to 0.92.
