Coal mining caused some subsided land,destroyed ecological en vironment and triggered some serious problems.Ecological restora tion of subsidence area of coal mines which was the main types of ecological restoration was one of the hot research topics in the fiel d of ecological restoration.The purpose was to improve the grow th and survival rate of plants in three types of solid wastes from coal mine.Two kinds of arbuscular mycorrhizal fungis,Glomus m osseae and Glomus versiforme were chosed to study the effect of pure fungi,mixed fungus on Physiological and biochemical char acteristics of Sorghum bicolor×S.sudanense in three types of coal mining wastes which were fly ash(S1),coal gangue(S2)and the mixture of fly ash and coal gangue(S3)and the mixture of sand and earth(S4)which was the control group.The results were su mmarizeed as follows:(1)The symbiotic associations were successfully established between AMF and Sorghum bicolor×S.sudanense.The inoculation of Glomus mosseae(G.m)can obviously improved the infection rate to 65.41%,68.72% and 58.00% in the substrates of S1,S3 and S4 and the mixed inoculation treatment had the best infection ra te to 67.14% in the substrate of S2.Show that two kinds of AM f ungis in the composite adversity has a strong tolerance.(2)The growth of Sorghum bicolor×S.sudanense was inhibited in the composite adversity of different types of coal mining waste s.The plant height,ground diameter,leaf area and dry weight c an significantly decreased and root length,root volume,root surf ace area,root average diameter,root activity and the uptake of N and P were inhibited.The colonization of AM fungi increased t he root length,root volume,root surface area of Sorghum bicolor×S.sudanense and the uptake of N and P by Sorghum bicolor×S.su danense to varying degrees,and the plant had different mycorrhi zal dependency in four substrates.The root average diameter si gnificantly decreased.Show that AM fungi exhibited the most obv ious role in promoting growth and nutrient uptake of Sorghum bic olor×S.sudanense grown in four substrates.G.m was more suitabl e for the revegetation of fly ash and the mixture of fly ash and c oal gangue,while G.v was more suitable for the revegetation of c oal gangue.(3)O2(·-) generation rate,H2O2 contentration and MDA content c an significantly increased and Sorghum bicolor×S.sudanense’s SO D,POD and CAT activities in three types of coal mining wastes w ere higher than those in S4.AMF inoculation can significantly enh anced POD and SOD activities and improved ASA content and O2(·-)generation rate,H2O2 contentration and MDA content can signific antly decreased.The result also showed that AMF inoculation mitigated the adverse effects on membrane organization and functio n,promoted plant grown and increased plant resistance of Sorgh um bicolor×S.sudanense via enhancing antioxidant enzyme activity and the ability to scavenge ROS in the composite adversity of di fferent types of coal mining wastes.AMF inoculation can improve the enzymatic defense system in Sorghum bicolor×S.sudanense a nd heighten its self-adjustment and adaptability to coal mine area s.(4)Sorghum bicolor×S.sudanense’s Soluble sugar content,solu ble protein content,proline content and the relative penetrability of cell membrane in three types of coal mining wastes were hig her than those in S4.AMF inoculation can significantly increased soluble sugar content,soluble protein content and the accumulati on of Proline and reduced the relative penetrability of cell mem brane.The result also showed that AMF had effectively mitigated the adverse effects of coal mining wastes on biological membra ne and the death of Sorghum bicolor×S.sudanense and improved p lant adaptation to strsss via increasing the content of osmoregul ation substance which contributed to increase the concentration of the cytoplasm,maintain a lower cell turgor pressure and the n ormal physiological metabolism and protect protein molecules fro m oxidative damage.(5)The photosynthesis of Sorghum bicolor×S.sudanense was i nhibited in the composite adversity of different types of coal mini ng wastes.The chlorophyll content,Car content,chlorophyll a/b,Fv/Fo,Fm,qP and Yield can significantly decreased,Foand NP Q can significantly increased.The colonization of AM fungi increased chlorophyll content,Fv/Fo,qP and Yield of Sorghum bicolor×S.sudanense and decreased Foand NPQ by Sorghum bicolor×S.su danense to varying degrees.The significant influence was not fo und on Fmbetween inoculation and uninoculated plants(P>0.05).AM fungi played a good role for the plant growth in four substrate s and was capable of alleviating the damage cased by the advers e factors of different types of coal mining wastes on Sorghum bic olor×S.sudanense by means of improve leaf chlorophyll content,p hotosynthesis,chlorophyll fluorescence and growth,and enhance Sorghum bicolor×S.sudanense’s stress-resistance. |
