Many kinds of diseases appeared in a lot of pig-farms in shandong province which brought great loss to our pig-rasing industrious in recent years, such as PRRS, PCV-2, M.hyopneumoniae, swine influenza. There are many pig-farms where over two diseases affecting the pigs in the same time. In order to make out the role of swine influenza in the agents causing pig diseases, primer of GP5 gene and primer of M gene were designed according to the sequences of porcine reproduetive and respiratory syndrome (PRRSV) and swine influenza virus(SIV)published in GENEBANK. 11 samples from sick and dead pigs from Shandong pig farms were detected for pathogenic porcine reproductive and respiratory ryndorme virus and swine influenza virus by RT-PCR, and 79 serum samples antibody against PRRS and SIV were detected by ELISA and HI.The results showed that the diagonasical method of PRRSV-GP5,PRRSV-GP6 and SIV-M was constructed by RT-PCR and sequence analysis . The PRRSV positive rate is 45.4% , the SIV positive rate is 0% by using the diagonasical method. ELISA for the detection of antibodies to PRRSV positive rate is 37.9%. HI for the detection of antibodies to SIV positive rate is 0%.Phylogenic analysis based on PRRSV-GP5 gene showed strain o Shandong close to branch with PRRSV isolates from Wuhan Weifang , PRRSV-GP6 gene close to branch with PRRSV isolates fromGuizhou.The results showed the length of complete M gene is 1027bp. The nucleotide homologies of the M genes of Tianjin strain belongs to the same branchu with HongKong and Guangdong.The nucleotide homologies of the NS genes between the tested strains and the reference strains were from 80.7% to 99.2%. Phylogenic analysis based on NS gene showed TJ2 and TJ4 strain belong to same branch with SIV isolates from Guangdong. Shanghai and HongKong, but sequence distance more close between tested strains and the SIV isolates from Guangdong and Shanghai than isolates from HongKong.
