This paper studies on the growth of tetraploid Eucommia and its cold resistance.And the main application components in the leaves of Eucommia and the gum are also studied.Some Eucommia plants are Screened out from the samples because their particular application,the specific results are as follows:1.In this study by reaserching the growth of diploid and tetraploid Eucommia samples,we ultimately determine whether growth of leaves or branches the growth the growth rate of tetraploid samples faster than that of diploid samples.This discovery has some significance to the current acreage of Eucommia less and more scarce raw materials;2.By the study of cold resistance of tetraploid Eucommia,we found the samples have different reaction.we found sample 6 has a relatively low conductivity Under low temperature stress,sample 8,9 and 19 have a higher levels of soluble protein,samples 9,10,17-18 have lower MDA content,meanwhile sample 11 has a most active POD.These samples all have a part of excellent cold tolerance and has a certain significance.3.By the screening of HPLC conditions of Chlorogenic acid,the final conditions as follows:Acetonitrile-0.4%phosphoric acid10:90,flow rate 1.0 mL/min;By the screening of extraction conditions of Chlorogenic acid,the ultimately optimal conditions were:drying temperature 60℃,70% ethanol solution,liquid ratio of 10,soaking for 24 h,ultrasound- assisted extraction time 60 min; in the above extraction conditions,chlorogenic acid is the maximum,less impurities,and had no significant effect;by research the chlorogenic acid in samples monthly,the best month to harvest chlorogenic acid is June;at last,sample 18 has the highst chlorogenic acid,samples 5,6,9,10,11,12,13,17 and 20 have higher chlorogenic acid,so could choose that several samples to a deeper research.4.By the screening of HPLC conditions of Aucubin,the final conditions as follows: Methanol-water88:12,flow rate 1.0 mL/min;By the screening of extraction conditions of Aucubin,the ultimately optimal conditions were: drying temperature 105℃,70% Methanol solution,liquid ratio of 16,soaking for 24 h,ultrasound-assisted extraction time 60min;in the above extraction conditions,Aucubin is the maximum,less impurities,and had no significant effect;by research the Aucubin in samples monthly,the best month to harvest Aucubin is June;at last,sample 16 has the highst Aucubin,samples 5,6,9,10and11have higherAucubin, so could choose that several samples to a deeper research.5.By the screening of HPLC conditions of Rutin,the final conditions as follows: Methanol-water35:65,flow rate 1.2 mL/min;By the screening of extraction conditions of Rutin,the ultimately optimal conditions were:drying temperature 60℃,60% ethanol solution,liquid ratio of 12,soaking for 24 h,ultrasound-assisted extraction time 50min;in the above extraction conditions,Rutin is the maximum,less impurities,and had no significant effect;by research the Rutin in samples monthly,the best month to harvest Rutin is July;at last,sample 6 and 15 have the highst Rutin,samples 5,8,11,13,16,17and 18 have higher Rutin,so could choose that several samples to a deeper research.6.By using the improved synthesis method,we determined the gutta percha in the samples.The results as follows:some of measured samples of the Tetraploid Eucommia was significantly higher than diploid samples;in these samples,sample 9 had a highest content, which pure rubber content was 7.37%.7.The average membership function this study selected tetraploid plants were larger than diploid plants,it concluded that tetraploid plants than diploid plants;In all tetraploid plants the average membership function 5,6,11,12,16,18 plants are better than other plants,while the 6 no significant difference between them. |
