Studies On The Routes Transmission Of Mice Infection With Porcine Circovirus Type 2(PCV-2)

The PCV-2 is the one of the causative disease which has risen humorous economic loss on swine industry. It's pathogeny is the PCV-2.The disease is characterized by Postweaning multisystemic wasting syndrome(PMWS). The object of the study is Kunming mice, The experiment is successful in Kunming mice infect the Porcine circovirus type2(PCV-2), observe the pathological changes of the system, For the production of life the spread of the Porcine circovirus approach provides proof.The study of vertical propagation,There are 3 steps for this experiment:inject the PCV-2 to the female mice when they pregneant 6-7days. This is the infection organism.The mice have the phenomenon of still birth and abortion. Cut down the tail and get the blood and do PCV-2 to confirm if the infection of PCV-2 is successful; Put the young mice into a experimental group.Use the Porcine circovirus type 2(PCV-2) infected female mice success, set the blank for comparing. Dissect its little mice when they are 0-day-old,14-days-old,28-days-old,49-days-old,56-days-old. The female mice have the phenomenon of still birth, but the young mice don't have clinical symptom during the detect days.After dissection we can find:clottednilk in the young mice's stomach who are 0 days, have PCV-2. The specificity of lactation rats have pulmonary hemorrhage disease hyperemia, Lymph node enlargement of bleeding, Histopathological observed on the lymphocyte necrosis and macrophage cell necrosis. PCR results shows that we can detect the PCV-2 in Lymph nodes, the thymus, thyroid, lungs, liver, brain, heart, kidneys, bowel, spleen, uterus and testicles at the age of 14-days-old,28-days-old,49-days-old. We can't detect PCV-2 in Lymph nodes at the age of 56-days-old.In contact with horizontal transmission, We put the healthy Kunming mice(no matter male or female) into the cage which successful infected the PCV-2 into the health Kunming mice, next day. After 40 days, they are all alive.Then they were dissected.The Porcine circovirus type2 (PCV-2) exist when PCR detected there nosedrops, saliva,urine, excrement. At the same time,the mice pulmonary hemorrhage, lymph node enlargement of bleeding.So we found that they can infect PCV-2 each other when they in same cage,In non-contact with horizontal transmission, group the cleaning Kunming mice randomly,5 mice were put in cage A, which infected PVC-2.Then we put 10 healthy mice in cage B, one meter from the cage A. After 50 days later, no mice died. Killed the mice from cage B, the pulmonary hemorrhage more obvious than lymph bleeding. The experimental results shows that PCV-2 spreads through the air, but The clinical symptoms is less than the former two ways of spreading.
