In this study, we used Bahiagrass mature seeds as explants to induce the callus granular,and then we used different concentrations (0 g / L,5 g / L,10 g / L,15 g / L,20 g / L) of NaCl as our selector,also we used the selection of Stepwise method,to filter out the salt-tolerant callus variantion and regenerate salt-tolerant plants.When the regenerate salt-tolerant plants were seeded survival after transplanting,we carried out with different concentrations of pouring salt treatment,we compared the physiological and biochemical indexes of Bahiagrass salt Callus culture, regenerate salt-tolerant plants and synchronization. The main conclusions were as follows:1 We founded that the germination rate of Bahia grass was increased with the NaCl concentration decreased significantly;also with the medium NaCl concentration increased, the callus induction rate, callus survival and regeneration rates and stability were significantly decreased。2 To deal with mature seeds of Bahiagrass granular salt induced callus and the kinds of general callus with different concentrations (0 g / L,5 g / L,10 g / L,15 g / L,20 g / L) of NaCl,The results showed that: With the increasing concentration of NaCl stress, there is an obvious difference in many ways between the Salt Group Bahiagrass callus and callus of the control group, the water content, MDA content of the former were Obviously higher than the latter;and the POD activity, SOD activity of the former were also higher than the latter. The study also found that the proline plays an important role in increaseing salt-tolerant of Bahiagrass calli.3 Poured the salt treatment on the regeneration and the physiological and biochemical characteristics of studies have shown that,with the improvement of pouring salt concentration,the Chlorophyll content and root activity of group of plant salt-tolerant variant and the general was first increased and then decreased ,and the group of plant salt-tolerant variant was higher than the general, the content of MDA of plant salt-tolerant variant and the general first rose and then fell flatlly. Membrane permeability of Salt-tolerant and control were both having a rising trend as the salt concentration increased, but the membrane permeability trend of salt-tolerant plants were significantly lower than the control which were both under the same conditions of salt stress.The variant were lower than the control under the same NaCl stress; The content of PRO in the variant were equal to the control without NaCl stress, in both the variant and the control, the content of PRO increased follow by the content of NaCl, the variant had higher accumulation of PRO than the control; The activity of POD, SOD increased first and then decreased on the whole both in the variant and the control, the variant were much higher than the control under the same NaCl stress.
