The study is to provide a experimental reference for the aerospace mutation breeding,using"SHIJIAN 8"satellite to carry eight type of Medicago seeds which is WL232,WL323HQ,BeZa87,Pleven6,LONGMU801,LONGMU803,ZHAODONG,CAOYUAN 1,and through the research of Medicago sativa L cytology effects,physiologic and biochemical index,seeding vigour and RAPD molecular engineering to bring out the results. The finding is as below:After the space mutation breeding,the eight type of Medicago sativa L seeds, WL232,WL323HQ,BeZa87,Pleven6,LONGMU801,LONGMU802,ZHAODONG,CAOYUAN 1 are showing a trend of rise in the mitotic index of root tip cells of SP1 generation,each increases 20.27%,19.53%,18.54%,12.73%,23.70%,27.03%,21.85%,12.28%. And the heterosomal of the eight type Medicago seeds also showed aberration ,the aberration rate is 5.12‰,9.76‰,9.78‰,15.26‰,14.36‰,4.45‰,3.73‰,7.99‰respectively,in which the chromosome fragment makes the largest percent.Measures the activities of peroxidase( POD) and Superoxide Dismutase(SOD),soluble protein content of the leaves of eight type Medicago sativa L. The result shows that, after the satellite"SHIJIAN 8"carrying,the activities of POD of the eight type Medicago sativa L's leaves each rise 8.02%,6.00%,15.64%,14.63%,17.12%,3.44%,13.64%,7.68%, soluble protein content of the leaves also increase 8.02%,6.00%,15.64%,14.63%,17.12%,3.44%,13.64%,7.68% respectively, and the activities of SOD all reduce each by 7.39%,2.78%,11.94%,6.42%,6.34%,4.03%,4.81%,12.47%.The data shows the the Medicago sativa L reduce damage from severe environment by increase POD activities,soluble protein content and reduce SOD activities.Measures the root length,the germination percent,the weight of the eight type Medicago sativa L,the result shows that,after the satellite"SHIJIAN 8"carrying,all Medicago seedlings are inhibited in root growing.The germination percent is variant from different type of Medicago, for example, CAOYUAN 1 and WL232 is higher than the control when Zhaodong,Longmu801,Longmu803,Pleven6,BeZa87 and WL323HQ is lower than the control.And the height of Zhaodong,Longmu801, Longmu803,Pleven6 and WL323HQ is lower than the control, respectively by 7.84%,2.94%,0.88%,4.06%,6.10%; when the height of Caoyuan1,BeZa87and WL323HQ is heighten each by 8.78%,8.47%,2.69% ; And comparing with the control,the weight of Zhaodong,Longmu801,Longmu803,Pleven6 and WL323HQ drop each by 6.52%,8,70%,2.27%,6.00%,6.82%,14.29%,12.27% ,when Caoyuan1 raise 8.33% specially.The study use 36 primers to excute RAPD-PCR to the eight type Medicago sativa L and their corresponding breed that the"SHIJIAN8"satellite,and select 21 primers to amplified clear,reproducible polymorphic products. Polymorphism of the products include increment and deletion. After the space flght,the eight type Medicago's RAPD Polymorphism is 18.06%,14.47%,7.04%,13.70%,15.00%,14.29%19.23%,11.39% respectively. And WL232 is the highest one in amplified polymorphic,which is 9.72%; and the highest deletion polymorphic is ZHAODONG,which is 12.82%,at the same time ZHAODONG is highest one in total polymorphism.
