Objective: To observe RECK and MMP-2 expression in normal oral mucosa(NOM),oral lichen planus(OLP) and oral squamous cell carcinoma(OSCC) and explore its significance in the incidence of oral cancer.Method: The western blot method were applied to detect the protein expression of RECK and MMP-2 in 17 cases with NOM, 27 cases of with OLP and 24 cases with OSCC.Results: The MMP-2 protein positive rate was 11.8(2/11),51.9%(14/27) and 91.7%(22/24) in NOM, OLP and OSCC group, respectively. The MMP-2 expression in cancer tissues was significantly higher than in NOM and OLP(P<0.05). The RECK protein expression in cancer tissues(4.2%, 1/24) was significantly lower than in NOM(94.1%,16/17) and in OLP(40.7%, 11/27). The MMP-2 expression rate was 36.4.5%(4/11) in tissues with positive(+) RECK expression. In tissues negative(-) for RECK expression,the MMP-2 expression rate was 62.5%(10/16). Pearson’s χ2test revealed that there was a significant negative correlation between the expression of RECK and MMP-2(x2=24.7,p<0.001) in OLP.Conclusion: Correlated with the development of oral lichen planus and malignant potential RECK and MMP-2 expression may be related. RECK expression rate of low malignant potential of oral lichen planus, on the contrary, oral cancer potential of MMP-2expression rate of the high expression of RECK and therefore can serve as a judge in oral cancer, one of the potential level of reference.
