Purpose:To study the purpura kang granule on BALB/c mice immune thrombocytopenic purpura(ITP) animal model of peripheral blood platelet, TPO, TGF- beta 1, IL-3, IL-6 and IL-11 level, the effect of preliminary discussion purpura kang granules treatment in mice and its possible mechanism of ITP.Method:Determination of SPF health BALB/c mice peripheral blood platelet count, mice were divided into normal group, model group, the blood beauty group, the prednisone acetate group, purpura kang high dose group, middle dose group and low dose group using the random number table, a total of seven groups, each group of 12, male and female half and half. The normal group, general breeding, do not do special treatment. The normal group and model group in experiment 1 d distilled water according to 0.5 ml / 20 g weight dose lavage, the rest of the various dosage groups in 1 d were given corresponding treatment respectively, one time a day, a total of 14 days. Compared with normal group, model group mice had depression injection of APS after 2 days, the colour were loose, cluster tiredly lay, activity weredecreased, food and the water to reduce; Groups of mice appeared obvious skin purpura, in the majority with abdomen, tail, and radical, model group hemorrhage phenomenon were the most serious. Compared with model group, the drug to alleviate after mice activities freely, improved mental state, automatic water and foraging, phenomenon of purpura were improved. 2 The influence of peripheral blood platelet count on the ITP model mice Compared with normal group, the peripheral blood platelet count of model group mice were falling significantly, P<0.05; Compared with model group, peripheral blood platelet count of each dose group mice were significantly elevated, P<0.05. 3 The influence of the peripheral blood TPO content on the ITP model mice Compared with normal group, TPO levels of model group were significantly decreased, P<0.05; Compared with model group, TPO levels of the blood beauty Ann group, purpura high dose group, middle dose group and low dose group were significantly increased, P<0.05. 4 The influence of the peripheral blood TGF- beta 1 content on the ITP model mice Compared with normal group, the content of TGF- beta 1 in model group had a marked increase, P<0.05; Compared with model group, TGF- beta 1 levels of the blood beauty Ann group, purpura high dose group, middle dose group and low dose group were significantly lower, P<0.05. 5 The influence of the peripheral blood IL-3, IL-6 and IL-11 content on the ITP model mice Compared with normal group, the peripheral blood IL- 3, IL- 6, IL- 11 concentration of model group mice increased significantly, P<0.05; Compared with model group, IL-3 concentration of traditional Chinese medicine(TCM) in the control group, high, middle dose group significantly lower, IL- 6, IL- 11 concentration of each dosage group decreased obviously, P<0.05.Conclusion:1 Purpura kang granule can effectively reduce bleeding in ITP model mice. 2 Purpura kang granules can relieve purpura and inflammation. 3 The treatment effect of dose group in Purpura kang granules in ITP is the better.
