Objective:To observe the NF-κBp50and IFN-y expression of mouse sinus mucosa of Acupuncture with Guanyuan moxibustion treatment of chronic rhinosinusitis(CRS) model, and explore the possible mechanism of Acupuncture with Guanyuan moxibustion treatment and the molecular biological development mechanisms of CRS.Method:160C57BL/6J mouses were fed one week after adaptation and randomly divided by DPS into normal group, model group, sham operation group, acupuncture group, western medicine group, traditional Chinese medicine group(regular doses), traditional Chinese medicine group(low-dose group), traditional Chinese medicine group(high-dose group),20in each group.(This paper only incorporates the datas of acupuncture group, model group, sham operation group, normal group). After CRS model established for8weeks, the acupuncture group were given needling at Zusanli and moxibustion on Guanyuan alternately and the western medicine group were given clarithromycin everyday. After14days, they were sacrificed to take sinus mucosa, NF-κBp50and IFN-y expression of sinus mucosa were detected by immunohistochemical method(S ABC method) and HE staining were observed by light microscopy.Result:1. Symptoms and physical signs:In model group, expressions of nasal obstruction, sneezing and nasal scratching always existed, accompanying by the symptoms of lachrymation and conjunctival congestion and edema, in acupuncture group and western medicine group, the frequencies of sneezing and nasal scratching decreased, and without lachrymation and conjunctival congestion and edem.2.HE section:in model group, the sinus mucosa showed chronic inflammation and mucosal infiltration of inflammatory cells, and epithelial cells were found, the array of cilia and epithelial cells were inordinate, even degenerative necrosis and exulceration,. In acupuncture group, sinus mucosa repaired obviously, and inflammatory cells and glands and goblet cell hyperplasia were not obvious.3.In model group, the mean optical density of NF-κBp50expression was significantly higher than the normal group and sham operation group(P<0.01); After treatment, the mean optical density of NF-κBp50expression of acupuncture group and western medicine group was significantly lower than the model group(P<0,01). In model group, the mean optical density of IFN-y expression was no difference by compared the normal group(P>0.05). After treatment, the mean optical density of IFN-γ expression of acupuncture group and western medicine group was significantly higher than the model group(P<0.05).Conclusion:The acupuncture with Guanyuan moxibustion adaped can effectively improve the body systemic symptom of the mouse model of CRS, decreasing the NF-K Bp50and increasing the IFN-y expression, inhibiting the release of inflammatory cytokines, correcting the Th1/Th2imbalance, improving the immune function of mouse modle of CRS, reduced the inflammatory reaction of sinus mucosa. Sinus mucosa repaired obviously caused by inflammation in this treatment.
