| Objective:This study to assess the effects of electro-acupuncture (EA) stimulation of auricular concha region (ACR) on the body weight, food intake and fat metabolism in obesity rats.Methods:604weeks SD male rats were housed in alone cages, randomly selected10as blank control group with normal breeding; than50feed with high-fat food for8weeks, Experimental studies in rats for10percent of body weight is higher than the average weight of the normal group,36building successful rats were randomly divided into model group, electro-acupuncture (EA) stimulation of auricular concha region (EA-ACR) group, shame electro-acupuncture (EA) stimulation of auricular concha region (sham-EA-ACR) group, n=12. Intervention:The normal group and model group were not stimulated; the rats ears of EA-ACR group each was placed two stimulating electrodes, ipsilateral electrodes are connected to the same stimulator, two electrodes were placed in the ear cavity and concha; reference EA-ACR group ear acupuncture stimulation, sham-EA-ACR group rats selected bilateral hip, ipsilateral placed two electrodes located on the sacrum and the greater trochanter of the femur in rats connection at the midpoint of this line lcm down position;Electroacupuncture parameters:2/15hz,2mA,30min/d, continuous electrical stimulation, daily for six weeks.After the intervention treatment, each group rats were weighed and recorded data, drawn by step and record the data and save samples. Elisa using in the serum norepinephrine (NE) content, using QPCR to research beta3adrenergic receptors and uncoupling protein-1exression in brown adipose tissue.Results:(1)compared with model group, EA-ACR group rats weight loss(P=0.02), epididymal fat reduction (P=0.02), brown fat content increased (P <0.05), No statistical difference in perirenal fat(P=0.31); Sham-EA-ACR group rats weight no significant decline (P=0.65), epididymal fat reduction is not obvious (P=0.47)brown fat content in statistics no difference (P>0.05), no statistical difference in perirenal fat (P=0.30); high-fat food intake of each group was significantly lower than the normal group (P<0.05),(2)no difference in high-fat food intake groups (P>0.05);(3)gastric emptying was no significant difference over the groups.(4)Compared with Sham-EA-ACR group, EA-ACR group rats weight loss(P<0.05), epididymal fat reduction (P=0.01), perirenal fat reduction(P<0.05),brown fat content increased(P<0.05).Conclusion: electro-acupuncture (EA) stimulation of auricular concha region(ACR) can produce weight loss*and its mechanism may be mainly through the promotion of fat metabolism.(5) serum Elisa showed that compared with Model group, EA-ACR group significantly increased on an empty stomach blood content of NE in (P=0.020); Sham EA-ACR group was not obviously increased blood NE (P=0.383).(6) QPCR shows that compared with Model group, EA-ACR group significantly increased shoulder of brown fat in beta3adrenergic receptor expression (P=0.000) and the expression of brown fat inside the UCP-1(P=0.000). Sham EA-ACR group cannot increase the beta3adrenergic receptors and the UCP-1(P=0.076) expression (P=0.890).Conclusion:EA-ACR can produce the result that reduce weight, its mechanism may mainly by activating brown tissue, promote fat metabolism. The adjustment role of food intake and gastric emptying was not found in the experiment. |
