Effects Of Moxibustion Tianshu And Zusanli On Expression Of MMP-1 And TIMP-1 In Colonic Tissues Of UC Rats

Purpose：The process of ulcerative colitis(UC)’s formation is related to thedegradation of extra cellular matrix(ECM).Matrix metalloproteinase-1(MMP-1)and tissue inhibitors of matrix metalloproteinase-1(TIMP-1) involved in theprocess of ECM’s degradation,and also influence the process of UC’s formation.The moxibustion therapy can stimulate the corresponding acupoints on the surfaceof the body and it has better adjustable function of the whole body, it alsohas a good prospect of application. This experiment observed the influence ofthe moxibustion on Tianshu(ST25)and Zusanli（ST36）points on the morphology ofthe rats’ colonic mucosa and the effects, and explore the regulating mechanismof the expression of MMP-1and TIMP-1in the UC rats’ colon tissue throughestablishing models of UC rats, to determine the moxibustion therapy cope withclinic and the comparable acupoints, and provide the more reasonableexperimental basis for clinical.Material and method：32male SD rats are randomly divided into blank group6rats and molding group26rats, kill2rats from the molding group after modeling.Divid the molding group into the model group, the Tianshu point moxibustion group,the Zusanli point moxibustion group and the western medicine group after modeling,each group has6rats. The TNBS/ethanol solution is used to occur the inflammationat the first time and DSS induce the inflammation disc-ntinuously. Mix5%TNBSand anhydrous ethanol into TNBS/ethanol solution. After getting the mixtureof5%DSS and distilled water, we inject100mg/kgTNBS mixture into the placewhich is about8cm above the ass of rats. Put the rats up side down for one ortwo minutes. On the seventh and the sixteenth day, we make the UC rat model byfeeding the rats with the solution of5%DSS. The each treating group is selectedTianshu point Zusanli point during the moxibustion therapy. Every acupoint have nine cones for each day, and have this treatment for fifteen days in all. Feedthe rats of the western medicine group sulfasalazine (SASP) by gastric gavageonce in each day, and also have this treatment for14days. Record the changeof the signs and symptoms of rats after treatment and observe the morphologicalchanges of colon tissues under the optic microscope and the electric microscope.Choosing the RT-PCR technique to detect and analyse the change of the biologicalindicators.Results：1. The state of mind of the UC rats in all the treatment groups are much betterthan that before treating. They drink and eat more, and more active. The weightsof the rats become normal. The frequency of bowel movement reduces without looseand bloody stools. The fur of the rats also becomes nice and clean.2. By visual inspection or observation through optical microscopy and electronmicroscopy, the colonic mucous membrane of all the treatment groups of UC ratsis much complete, and the glands arrange regularly. The recovery of the colonicmucous membranes’ damage is much better. The forms of the colonic mucousmembranes are close to the blank groups, and are superior to the model ones.Among all the treatment groups, the recoveries of the colonic mucous membranes’damage in the Tianshu point moxibustion group and the western medicine groupare superior to Zusanli point one.3. Comparing the expressions of MMP-1and TIMP-1of the UC rats’ colonic mucosain each group(chart2): The comparisons between the treatment groups and blankgroup show the expressions of two indexes have the upward trend,and thedifference between them is significant (P<0.01). The comparisons between thetreatment groups and model group mean that the expressions of two indexes havethe downward trend which means the difference between them is observable(P<0.01).The comparisons of all the treatment groups veal that the expressions of twoindexes in the Tianshu point moxibustion group and the weatern medicine groupare lower than that of the Zusanli point moxibustion group, and the difference between all the treatment groups is remarkable(P<0.01),except the differencebetween the Tianshu point moxibustion group and the weatern medicinegroup(P>0.05).Conclusion：1. The moxibustion on both Tianshu and Zusanli point can improve thesymptoms,signs and colon tissue morphology of UC rats.The effect of Tianshu pointmoxibustion group is better than that of the Zusanli point moxibustion group.2. The therapeutic effect of moxibustion on UC can be achieved by controllingthe expression of MMP-1and TIMP-1.
