Objective:This study is based on early experimental research, to discuss mechanism of Du Yiwei against to fibrosis through the observation effects of Du Yiwei particles on hepatic tissue expression of TGF-β1in hepatic fibrosis model rats, to expand the clinical application of Du Yiwei and provide theoretical support for clinical.Methods:78male SD rats are randomly divided into6groups (each group has13rats), including blank control group, Du Yiwei particles (high, medium and low dose) group, positive drug group, model group. To make fibrosis animal models with compound of CCl4hypodermic injection and high-fat diet while rats of blank control group receive continuously olive oil injection once per three days for8weeks. From the first day the experiment began, positive medicine group was given colchicine (0.045g/kg)gavage, each treatment group of Du Yiwei was given high(2.4g/kg), medium(1.2g/kg)and low dose(0.6g/kg)of particles gavage while model and blank group were given equivalent normal saline for8weeks. When the experiment ended, all rats were be executed with femoral artery bleeding, and then to test the contents of hyaluronic acid (HA), laminin(LN),procollagen type Ⅲ(PCⅢ) and collagen-Ⅳ(Ⅳ-C) in the serum with radioimmunoassay. To observe the form of organization and fibroplasia with optical microscope after HE and Masson staining and to test the TGF-β1protein express and mRNA express with immunohistochemical method and real time PCR method.Results:①The serum indexes (HA、LN、PCⅢ、Ⅳ-C contents):Compared with blank control group, hepatic fiber index of model group rats were significantly elevated(p <0.01); Compared with model group, serum indexes in each dose treatment group were significantly induced(p<0.01); Compared with positive medicine group, have statistically significant difference were observed in the treatment group.②The pathological morphology test suggested that compared with the blank group, model group rats showed hepatic lobule structure damages, inflammatory cells infiltration. Rats of positive medicine group showed basically normal lobular architecture, only see a small amount of collagen deposition portal area; each dose group of Du Yiwei showed exist of hepatic lobule structure but not in alignment, a small amount of inflammatory cells infiltration were visible.③Immuno-histochemical test results TGF-β1in hepatic tissues of rats:a small amount of TGF-Pi basic expression were visible in the hepatic tissue stromal cells of blank group rats; model group rats showed TGF-β1expression significantly increased and distributed in brown cytosolic; rats of each dose group showed a small amount of TGF-β1basic expression in portal area and fiber interval. However, there is no statistically difference in the comparison between model group and positive medicine group.④TGF-β1mRNA expression level in hepatic tissues:TGF-β1mRNA expression in model group was significantly higher than blank control group(p<0.01); rats in each dose group showed statistically difference when compared with model group (p<0.01); Results compared with colchicine group, high dose group showed statistically significant difference when compared with positive medicine group(p<0.05).Conclusion:Du Yiwei particles can significantly reduce main serum hepatic fiber indexes of (HA, LN,PCⅢ,Ⅳ-C) contents in hepatic fibrosis model rats, reduce the pathological change of hepatic tissue, effectively restrain the expression of TGF-β1, results of the study suggested that Du Yiwei has good effects of anti hepatic fibrosis.
