Objective: To investigate the effects of Danshensu on activation and proliferation of HSC and on the expression of transforming growth factor beta receptor typeⅠandⅡin HSC,and to elucidate the anti-fibrotic molecular mechanisms of Danshensu.Method: The rat HSC was isolated with collagenase in situ-liver recirculation perfusion and cultured in vitro. MTT colorimetric assay was used to detect proliferation of HSC treated with different concentration of Danshensu. The expression ofα-SMA and TβRⅠ,Ⅱwas observed by immunocytochemical or indirect immunofluorescent staining and western blotting when TGF-β1-stimulated HSC was incubated with different concentration of Danshensu for 24 hours.Results: Danshensu had a dose-dependent inhibiting effect on the activation and proliferation of HSC.The eligible working concentration on HSC of Danshensu range from 0.15mmol/L~0.30mmol/L. Danshensu could down-regulateα-SMA and TβRⅠ,Ⅱprotein expression(P<0.05 or P<0.01) in TGF-β1-stimulated HSC,these effects are correlated with concentration.Conclusion: Danshensu could inhibit the activation and proliferation of HSC through down-regulating the expression of TβRⅠ,Ⅱlocated in cellular membrane of HSC. It might be the mechanism of Danshensu against hepatic fibrosis.
