Comprehensive learning of non-alcoholic steatohepatitis (NASH)in both modern medicine and traditional Chinese medicine(TCM)is demonstrated through literature review and experimental study in this paper. We discussed the efficacy and mechanism of warming phlegm and retainied fluid, cleaning heat and promoting dieresis to intervent nonalcoholic steatohepatitis. This research may give a great contribution to clinical therapeutic levels and new grug discovery.1 literature reviewThrough literature analysis in recent 10 years,we summarized the causes of NASH, the treatment of modern medicine, pathogenesis. And also researched status of Chinese medicine on the etiology and pathogenesis of NASH, Syndrome characteristics, research progress in clinical research, medical treatment, etc.2 Experimental study of NASH rats induced by high-fat diet by the way of warming phlegm and retainied fluid,cleaning heat and promoting dieresisObjective:To investigate the efficacy and mechanism of warming phlegm and retainied fluid, cleaning heat and promoting dieresis on rat model of NASH.Method:High-fat diet-induced rat model of NASH, and drug intervened by Yinchenhao Decotion, Lingguizhugan Decotion, Yinchenhao and lingguizhugan Decotiong while in modeling;The change of serum ALT, AST, TC, TG, HDL-C, LDL-C,IR,and the liver tissue DGAT2, PKC ε of Western-blot detection had been tested to observe the efficacy, Meanwhile,we observed the oil red 0 staining of liver biopsy.Outcome:(1) General conditions and pathomorphology:The hepatic steatosis and inflammation were increased in model group, which were improved in the treatment groups,. And there was no significant difference between treatment groups.(2) Serum transaminase:model group, serum ALT, AST levels were significantly higher than the control group, there is a statistically significant difference (P<0.05), The serum ALT, AST levels of Yinchenhao Decotion group.Lingguizhugan Decotion group, Yinchenhao and Lingguizhugan Decotion group were significantly lower than those in the untreated group (P <0.05).(3) Lipid:Compared with the control group, the model group was significantly lower serum HDL, serum TC, TG, LDL significantly increased (P<0.05). Yinchenhao Decotion group and rosiglitazone compared with the model group, serum TG statistically significant (p<0.05); Yinchenhao Decotion group,Lingguizhugan Decotion group, Yinchenhao and Lingguizhugan Decotion group serum LDL Compared with the model group decreased significantly (P<0.05), while serum HDL increased significantly (P<0.05). Those group compared with the rosiglitazone group, the difference was statistically significant (P<0.05).(4) Insulin resistance:Compared with the control group, IR value of the model group increased each treatment group relative to the untreated group could make it lower, with statistical significance (P<0.05) compared with the model group.(5) Liver tissue DGAT2, PKCε Western-blot expression:Compared with the control group, model group, liver tissue DGAT2, PKCε Western-blot expression were increased (P<0.05); for DGAT2, Yinchenhao Decotion group, Lingguigan and Yinchenhao Decotion group, rosiglitazone group compared with the model group was statistically significant (P<0.05); for PKC s, Yinchenhao Decotion group Lingguizhugan and Yinchenhao decoction group compared with the model group was statistically significance (P<0.05).Conclusion:The way of warming phlegm and retainied fluid, cleaning heat and promoting dieresis can significantly improve liver pathology of the high-fat diet-induced NASH rats;We could improve steatosis reducing serum transaminase levels; improving dyslipidemia; and can reach the intervention effect by inhibiting the expression of DGAT2, PKCε’s.
