Object Observed the growth and metastasis of subcutaneous tumor in mice, to evaluate the effect of the icariin and wogonin on the lung cancer associated with depression model. And explore the mechanism of icariin and wogonin by monitoring the expression of inflammatory cytokines, angiogenesis and tumor metastasis related factors and other factors.Method1. Established the model of social stress depression, and monitored the behavior change in mice by using the Noldus behavior monitoring system.2. Established several different modes of the lung cancer associated with depression model, and monitored the behavior of mice by Noldus behavior monitoring system. Detected the expression of VEGF in the serum of mice by Elisa; detected the expression of pERK, mmp-2, mmp-9in the lung tissues of mice by Western blot; detected the expression of VEGFR2mRNA and LI CAM mRNA in the lung tissues in mice of different groups by R-T PCR.3. Established the social defeat model of depression, and interfered with icariin and icaritin. Detected the expression of IL-6by Elisa, and detected the expression of BDNF mRNA, p-TrkB mRNA by Real-time PCR.4. Established a model of lung cancer after social defeat stress, and interfered with icariin and wogonin, detected the amount of VEGF, mmp-2and mmp-9which were associated with tumor growth and metastasis, and the expression of IL-6which were related to the inflammation.Result1. In the model of social stress depression, when compared the behavior of mice between control group and social stress group, some depressive behaviors such as less exercise and avoidance behavior could be observed in mice of the social stress model group.2. The subcutaneous tumor in mice of the social stress before tumor inoculation group grew fastest, and the metastasis of the tumor was happened earlier than other groups.3. Icariin and icaritin could improve the depressive behavior of mice by reducing pro-inflammatory cytokines and increasing the expression of BDNF.4. The icariin and wogonin could inhibit the growth and metastasis of subcutaneous tumors in mice, and reduced the expression of inflammatory cytokines, and tumor growth and metastasis related factors.Conclusion Social stress could promote the growth and metastasis of subcutaneous tumor in mice, and among the different modes of the lung cancer associated with social stress model, the subcutaneous tumor in mice of the social stress before tumor inoculation group grew fastest, and the metastasis of the tumor was happened earlier than any other groups. Icariin could improve the depressive behavior in mice, and wogonin could inhibit the inflammatory of depressive mouse.
