Objective: the purpose of this issue are through two parts of experimental andclinical investigation, in the hope of to find out the relationship between KneeOsteoarthritis and the hip joint’s degeneration.Method:The experimental part: in the first place,to establish a rat knee osteoarthritismodel which similar to human knee osteoarthritis’s pathogenesis, namely, the doubleforelegs cut induced the rats stand-up combined with Hulth model.in the secondplace.based on this model, By HE staining,toluidine blue staining, the red solid greenstaining, CollagenⅡ,MMP-13,Aggrecan,CollagenⅡ immunohistochemical staining.Inorder to study the relationship between Knee Osteoarthritis and the hip joint’sdegeneration.in the third place.through drug intervention, to observe the effect ofGubi mixture to cut the double forelegs induce the rats stand-up combined with Hulthmodel.Finally.The clinical part: to investigate the standard of122cases of patientswith knee osteoarthritis and fill in the questionnaire, using SPSS17.0statisticalsoftware to analysis the statistic was performed on the results, and draws the conclu-sion.Results: Firstly,through the HE staining,toluidine blue staining, the red solid greenstaining, CollagenⅡand MMP-13immunohistochemical staining, We can see in thedouble forelegs cut induce the rats stand-up combined with Hulth model in which thedegeneration of Knee joint is worest,the percentage of CollagenII positive cells is thelowest, the percentage of MMP-13positive cells and the Mankin’s score is the high-est combined with other groups.Secondly,through the HE staining,toluidine bluestaining, the red solid green staining, Aggrecan and CollagenⅡ immunohistochemicalstaining, We can find that the double forelegs cut induced the rats stand-up combinedwith Hulth model in which the degeneration of hip joint is worse,the percentage ofCollagenII positive cells is lower, the percentage of MMP-13positive cells and theMankin’s score is higher combined with the standing group.Thirdly,through the HEstaining,toluidine blue staining, the red solid green staining, CollagenⅡand MMP-13immunohistochemical staining, We know that the Gubi mixture’s model in which thedegeneration of Knee joint is lighter,the percentage of CollagenII positive cells is the higher, the percentage of MMP-13positive cells and the Mankin’s score is the lowestcombined with other groups.Finally,In KOA patients, knee joint’s pain, morningstiffness, limitation of activity and X-ray examination has no correlation with the hipjoint’s pain, morning stiffness, limitation of activity and X-ray examination.Conclusion:From what has been discussed above, we may safely draw the conclusionthat:First of all,we succeed established the different models about the rat kneeosteoarthritis, and the double forelegs cut induced the rats stand-up combined withHulth model is the most typical which is similar to human knee osteoarthritis’spathogenesis.in the next place,the double forelegs cut induced the rats stand-upcombined with Hulth model in which the degeneration of hip joint is worse,The Ratswith knee osteoarthritis which can aggravate the hip joint degeneration.Thirdly,Gubimixture accords with the regularity of traditional Chinese medicine in the treatment ofknee osteoarthritis in recent10years, Gubi mixture can delay the degeneration ofarticular cartilage of knee joint.In the end,In KOA patients, knee joint’s pain, morningstiffness, limitation of activity and X-ray examination has no correlation with the hipjoint’s pain, morning stiffness, limitation of activity and X-ray examination, so wecan come to the conclusion that the knee osteoarthritis has no correlation with thehip joint’s degeneration.
