| The stem segments of Loropetalum chinense var.rubrum were used as explants in study of organ culture to establish a rapid technique system of micro propagation, and to study the effects of leaf color in tissue culture. By means of the orthogonal experiment design, analysis of variance and multiple comparisons, the best medium and culture procedure of each phase were analyzed. The results were as follows:1) The best explants were stem segments which were easier to be sterilized by 0.1% HgCl2 in 8 min. MS+BA1.0mg/L+IBA0.05mg/L was the best initiation medium, and the inducement rate could be 74.0% after 30 days.2) The middle part in the same stem was the best part of explants, which had low contaminated rate and better sprouting effect.3) June was the best period of picking the explants and had the best sprouting effect.4) Improved MS+BA1.5mg/L+KT0.1mg/L+IBA0.4mg/L was the best medium of multiplication of clustered buds, and the multiplication rate could be 4.77.5) The pigment content in the leaves was influenced by illumination intensity. Along with the increased of illumination intensity, then the content of chlorophyll decreased and the content of anthocyanin increased gradually. It was the best condition of culture, which condition with 15001x of illumination intensity, 12h of light time and 20℃.6) Along with the increased of the content of BA, then the content of anthocyanin decreased gradually.7) Sucrose was the important factor determining the content of anthocyanin. Along with the increased of the consistency of sucrose, then the content of anthocyanin increased gradually. The best medium of multiplication containing 40g/L sucrose was better. |
