In order to establish leaves in vitro regeneration system for Loropetalum chinense var.rubrum, choose four representative varieties of’changyeMeihong’, "NO. 2",’Nenyehong’,’Dayehong’ as the experimental materials, the crucial effects of light quailities and plant growth regulators and sucrose concentration on callus induction and plant regeneration in Loropetalum chinense var.rubrum. Besides, studies of leaves in vitro culture process analysis of anthocyanin and the related physiological and biochemical index changes. The main research contents and results were as follows:1.The Loropetalum chinense var.rubrum leaves in vitro culture and plantlet regeneration.A suitable medium for leaves blade callus induction is MS+NAA1.0 mg/L 2.0+6-BA0.5 mg/L, Callus obtained can be used as materials of callus redifferentiation. Under the different light quality the callus induction rate is different, the red light is the best and blue light is the worst. The best medium of callus induction containing 30g/L sucrose was better.The L.chinense var.rubrum ’changyeMeihong’ optimal hormone combination for regeneration is TDZ 2.0mg·L-1+6-BA 1.0 mg/L+IBA 0.8 mg/L. The "NO.2" and optimal hormone combination for regeneration is TDZ 2.0 mg/L+6-BA 1.0 mg/L+IBA 0.5 mg/L. The L.chinense var.rubrum "NO.2" and L.chinense var.rubrum ’Nenyehong’ optimal hormone combination for regeneration is TDZ 2.0 mg/L+6-BA 1.0 mg/L+IBA 0.8 mg/L, The L.chinense var.rubrum ’Dayehong’ optimal hormone combination for regeneration is TDZ 1.0 mg/L+6-BA 1.0 mg/L+IBA0.5 mg/L. The results showed the regeneration efficiency of leaf is different,the highest one was in L.chinense var.rubrum ’Nenyehong’.It could has a higher regeneration efficiency when the leaves with the treatment by white light and the best medium of callus induction containing 40g/L sucrose was better.2. The determination of physiological and biochemical indexes in the process of Loropetalum chinense var.rubrum leaves in plantlet regeneration.Obtained by NAA induced callus, anthocyanin and soluble sugar content, and high content of soluble protein, POD, SOD activity,but obtained by picloram and 2,4-D inducing callus content is just the opposite. The regeneration cycle of soluble sugar content changes as follows:to reduce and to rise and to reduce; Soluble protein content changes as follows:to reduce and to rise and to reduce and to rise and to reduce; POD activity change as follows:to rise and to reduce; SOD activity change as follows:to rise and to reduce and to rise and to reduce; CAT activity changes as:to reduce and to rise. The physiological and biochemical indexes change trend similar under different light quality,but its content and activity of the cultivation of the peak and the peak time is different.
