Loropetalum chinense var.rubrum belongs to Hamamelidaceae,evergreen shrubs or sub-tree.It has beautiful red flowers and bright leaves,aspect change obviously,which is a species of precious ornamental plant originating from Hunan province.This research include the growth and differentiation effect of colchicines on different materials from Loropetalum chinense var.rubrum by the soaking method and colchicines mixed in culture.In view of vitrification in vitro,morphological, physiological characteristics,the forming factors of its vitrification have been studied. In order to reduce the vitrification,which is the base of Loropetalum chinense var. rubrum's commercial production.The main results were shown as follows:1.Abnormal growing phenomenon of stem apex is tricuspidate leave,slow growing,vitrification,callus is browning,colchicines on callus with soaking method like paste shape,and clump shoots is dwarf,larger stems,thick leave.2.In order to reduce the percentage Of death,vitrification,The optimum method is clump shoot with colchicines mixed in culture by 0.005~0.02%for 10~20 days.3.Loropetalum chinense var.rubrum vitrification shows the tow form:One shows that slight stem,leave growing faster,the length of leave is twice as normal; Another shows that lotiform leave,dwarf,easy ecclasis.4.The difference of the histiocytic observation by paraffin section between the normal shoots and vitreous shoots was carried in Loropetalum chinense var.rubrum. The results indicated that the epidermic cells arranged loose;the arrangement of the spongy tissue was irregular,the intercellular space was more broader,and the cytoplasm was more washy;the arrangement of the palisade tissue also decrease;the vascular bundle sheath also arranged loose,the inner diameter was amplification.5.The comparisons of the physiologic and biochemical difference between the normal shoots and vitreous shoots were carried out in Loropetalum chinense var. rubrum.The results showed that total Chlorophyll,anthocyanin content activity decreased,the water content,SOD and POD activity increased.6.Considering the balance of vitrification and multiplication,adding 0.5~1.0mg·L-16-BA,0.1~0.5mg·L-1NAA,1.2%agaragar,could suppressed vitrifying effectively.Adding 0.1%AC or 1g·L-1 PVA,could got the lowest rate of vitrification; The effect of vitrification on 2%~6%sugar is inapparent.The optimum medium in orthogonal experiment is:MS+6-BA1.0 mg·L-1+sugar3%+agaragar1.2%+AC0.1% or PVA 1g·L-1.7.Effect on different explant of vitrification rate is obvious.Comparison of stem apex,the meddle ministry stem and the foundation ministry stem,The obtained results showed that vitrification rate stem apex>meddile ministry stem>the foundation ministry stem.In addition,reducing subculture time to control the vitrification rate.
