| Six strains of Trichoderma in the laboratory were identified. The result showed that. T-33 and T-13.T-6 and T-22,T-34, T-1 were Trichoderma aureoviride, T. harzia(?)m, T. longihrachia(?)um and T.koningii respectively. Chlamydospore productions in different culture time were compared under the same condition. Chlamydospore sporulation of T-33 was fast. The yield of chlamydospore was highest. The fermentation, germination, storage time and effects on seed germination and damping-off of capsicum were studied with the T-33.The research on the conditions of chlamydospore production for the liquid fermentation showed that, the better single factors were oat liquid media, 30℃, 120r/min surge culture, sucrose as carbon source, (NH4)2SO4 as nitrogen source and the ratio of 20 to 1 for C/N. Under the better single factors mentioned above, the largest quantity of chlamydospore, i.e.3.05×107spores/ml, was obtained on the 10th day after culture. In order to obtain the best combination of the important single factors, temperature, pH and surge speed, which possessed the more important effect than others, the orthogonal test was made. The result showed that the combination of 30℃, pH4 and 140r/min was best, the yield of chlamydospore was 3.37×107 spores/ml.The germination of chlamydospore under the different conditions was tested with the method of water agar strip on the slide. The effects of light, spore mature condition and liquid media on the germination were not different significant, more than 80% of the germination were made at each treatment of these factors 8 hours later. However, the effects of different temperature, pH and relative humidity were quite different. The chlamydospore could germinate best respectively at 30℃, pH4 and RH100%, the germination rates were 99.4 %, 98.63% and 68.89%, respectively.The storage experiment of chlamydospore indicated that, in each gram of chlamydospore powder and mixture of chlamydospore powder and talcum powder, the quantity of chlamydospore decreased with the storage time increasing, the germination rate of chlamydospore also decreased to some extent. The number of pure chlamydospore powder stored at 4℃ was 2.1×108 spores/g 8 months latter, and chlamydospore still had 68.01% of germination rate; However, the number of the mixture stored at 25℃ decreased at 3×107 spores/g, and germination rate was just 38.04%. Therefore, the chlamydospore of Trichoderma could survive a longer time.The ferment filtrate obtained when chlamydospore were produced with the method of liquid fermentation influenced the seed germination of capsicum greatly. |
