Pochonia chlamydosporia is an important biocontrol fungi.The fungus can be used to control not only plant parasitic nematodes but also animal parasitic nematodes.It has great potential to control parasitic nematodes.In order to obtain more information about P.chlamydosporia ARSEF3539.Its morphological characteristics,fermentation broth and it s effect on animal parasitic nematodes were studied.The results are listed as follows:The colony morphology growth rate of the fungus grew on the 0.4 g/L corn-meal-agar medium was slightly different with that grew on Savary-gilliard glucose agar medium.The colony radius was 2.4 cm on CMA and 3.0 cm on SDA,respectively.The hyphae on the two medium were both radial,thick,and developed.Conidiophores were single or verticillate born in hyphae.Each conidiophore had 4~5 branches.and there was an oval or round transparent conidium on the top of each branch.Chlamydospore with thick wall was formed in the aged hyphae and each was brick lattice shape and multicellular.The Caenorhabditis eleguans and eggs,Strongylus larvae and eggs.Ascaris suum eggs were put on the CMA medium surface with the fungus.The results showed that the infection rate of the fungus to C.elegans eggs and Strongylus eggs was only 2%;The fungus can break the eggshell and infect the eggs,and the infection rate was 29.6%after one week.Four different liquid medium were used to obtain fermentation broth of P.chlamydosporia ARSEF3539.The fermentation broth filtrations were collected,and the protein content.enzyme activity,phosphatase activity and so on were determined.The results showed that the indicators of the fermentation broth were also different in different medium.The protein content and enzyme activity of LMZ medium,which also had the highest inhibition rate to the tested nematodes,was the highest among the four and the corn was the lowest one.Through the study,the morpholog,ical characteristics,the effect on animal parasitic nematodes and growth in four different liquid medium of P.chlamydosporia ARSFEF3539 were determined,and useful information from the analysis of the results could be supplied to the storage of P.chlamydosporia,the researches on the mechanism of the prevention and control by the biological agents. |
