Dysosma versipellis (Hance) M.Cheng, which belongs to the Berberidaceae, is a medicinal plant with several natural compounds isolated form it. Podophyllotoxin is such a compound currently investigated for carinogenicity, cytotoxcity and antitumor activity. In recent years, with increasing demand for anti-tumor drugs, D. versioellis has been over-collected and is meeting with imminent danger. In order to protect and utilize this species, we firstly reported the propagation system of D. versipellis and explored the method of enhancing the content of podophyllotoxin. The main results were as follows:1) Direct shoot regeneration was achieved by culturing 1.5cm X 1.5cm rhizomes on MS medium enriched with 1mg/L BA and 0.2mg/L TDZ, the inducing rate is 100%. The rhizomes expanded 3 or 4 times after 20d, then were cut the size of 1.5cm X 1.5cm and planted optimum propagation medium is MS + BA 0. 5mg/L + NAA 0. 2mg/L,with propagation ratio of 8-10 after 4 weeks culture.2) The medium MS+NAA 1mg/L+KT 0.2mg/L or MS+NAA lmg/L + BA 0.2mg/L is suitable for inducing callus with root explants, the callus is light yellow, loose, pellet which regenerated roots; The medium MS+TDZ 0.2mg/L+NAA 0.1mg/L is suitable for inducing callus with stem explants, the callus is green, compact, lump which regenerate shoots.3) The rooting medium is 1/2MS + NAA 0. 5mg/L or 1/2MS + IBA 0.5 mg/L, rooting rate is 100%, AC is useful for rooting, the best concentration is 0.05%.4) Content of podophyllotoxin was determined by HPLC and compared different parts of cultured D. versipellis (leaf, stem rhizome, root), the result is that podophyllotoxin...
