| This study is designed to investigate the anti-abortive effect and mechanism of Scutellaria baicalensis Georgi and it's ingredients Baicalin, Baicalein on uterus immunity in Mifepristone-induced and Lipopolysaccharide induced abortion models.The function of RU486 and LPS inducing mouse abortion rate is Compared between the two models and the anti-abortive action of Scutellaria baicalensis Georgi observed. Micro and super-micro changes of the uterus tissue are analysed. The contents of cytokine IFN-γ, IL-2 and IL-4, IL-10 in uterus lysate are measured;The express ratio of costimulatory molecules CD80 and CD86 on spleen cells is studied;the content of progesterone in uterus lysate is assayed, in order to explore the mechanism of scutellaria baicalensis Georgi regulating Th1/Th2 immune balance and hormone during embryo implantation.Ten-week-old BALB/c mice are divided into 8 groups randomly. A as control group, B as RU486 model group, C as anti RU486 group, D as LPS model group, Group E used PXF anti LPS, then else Group FB,FD used Baicalin, Group GB,GD used Baicalein and group HB,HD used scutellaria baicalensis Georgi. RU486 as B or LPS as D is Injected on day 7 of gestation to induce abortion. Meantime anti-abortive herbs or ingredients are used. On day 9 of gestation the mice are sacrificed by cervical dislocation. The rate of abortion, uterus tissue micro- and super-micro changes are observed. The contents of cytokines IFN-γ, IL-2, IL-4, IL-10 and P4 in uterus lysate are assayed by ELISA, The express ratio of costimulatory molecule CD80 and CD86 on spleen cells is flowcytometried .The Abortive rate of Groups B,D is 81.2%, 80.0% and the rate of embryo absorption is 86.4%, 85.7% respectively. The difference is significant compared with the control(p<0.01). In Groups B,D the uterus tissue is observed and has obvious inflammation, damnification at behind of embryo development. Super-micro structure has some changes such as mitochondrion dead, more lysosome, less secrete granules. P4 of uterus in group B is higher because RU486 blockes the combination of P4 and progesterone receptor, and resultes in P4 biological function loss and incretion imbalance. RU486 descendes the levels of IL-2, IL-4, IL-10 just as inhibiting immunity and increases IFN-γlevel in uterus, so Th1/Th2 immune balance leans to Th1 harmful response of gestation. LPS induces IFN-γ significantly, has no obvious effect on IL-2, IL-4, IL-10, increases Th1 immunity, destructes the balanceof Thl and Th2, and leads to gestation failure. Results show that the ratio of CD80 and CD86 had the same trend with that of IL-4/IFN-ywhich represents Thl and Th2 immune balance.Scutellaria baicalensis Georgi and it's ingredients: Baicalin, Baicalein can inhibit efficiently the abortions caused by RU486, LPS, and makes the abortive rate below 40%, close to normal level. Groups FB,GB,HB increases the levels of IL-2, IL-4, IL-10 in uterus contrast RU486, regulated to Th2 dominant and inhibites RU486 competition with P4. Scutellaria baicalensis Georgi and it's ingredients: Baicalin, Baicalein could descend IFN-ylevel caused by LPS, at the same time enhances the levels of IL-4, IL-10 in uterus. This is to say that inhibiting Thl and enhancing Th2 has double actions on Thl/Th2 immune balance disturbed by LPS, creating suitable environment for embryo development. |
