Prunus Persica was one kind of important fruit trees in the world. Peach used to be warmly received by the consumers and it played a great role in the fruit markets because of its beautiful color, sweet smiling and good quantities. But peach trees were badly harmed by virus. China was the biggest peach production country. The virus diseases also did a huge loss in peach production and limit the development of peach trees. Now in the world, with the development of modern biology techniques, tissue culture was mainly used for virus-free seedling research.In this paper, stem segments with bud of peach variety--white Chuznzhongdao peach were used forexplants. The variety was imported from Japan in 1991 by Liu Hanyun, vice professor of the Forestry Institute in Shanxi Agriculture University. The best media of white Chuznzhongdao peach in vitro culture and its culture procedure were mainly studied. The results were as follows:Primary culture medium: MS + BA 1.0 mg/L + IBA0.1 mg/L. The explants were effectively treated by NaClO (0.5%) for 20 min. Mid January was the best sampling season. In order to prevent pollution, 500ppm streptomycin or gentamycin sulfate could be used effectively.Subculture medium: MS+ BA 1.5 mg/L + IBA 0.2 mg/L. The propagation rate could reach about 7.33 for 30 days. After 5 times, the induction and differentiation came back normally, replacing MS by modified MS.Rooting medium: 1/2MS + IBA 0.5 - 1.0 mg/L + 0.1 %AC. 20 days later, the average of roots was up to 3.73-5.40, the rooting rate was 29.8% ~ 40.3%, the shortest rooting time is after 13.3 days.The highest survival rate of transplanted tube-seedling was 52.0%. The seedlings were transplanted in substratum made of sand and pearl ite (1:1).The purpose was to establish the tissue culture technique system of white Chuznzhongdao peach, providing for next productions of virus-free peach seedlings.
