Abstract/Summary:
The biochemical markers was done by the PAGE of CytochromeOxidas(eCOD)isozyme and the SDS-PAGE electrophoretogram of all protein in four isonuclear alloplasmic male-sterile lines, the restoring lines and the corresponding maintainer line of NIAN type non-1BL/1RS wheat; with microscopic examination about the development of pollen of male-sterile lines, the PAGE of COD isozyme was used to research the expression regular of COD isozyme in four isonuclear alloplasmic male-sterile lines, the restoring lines and the F1, at the same time, the SDS-PAGE about all protain in the development of pollen of eight isonuclear alloplasmic and allonuclear isonuclear male-sterile lines was used too. The results were as follows: 1. The isozyme patterns of COD can be used to identify the four isonuclear alloplasmic male-sterile lines,the maintainer line and the restoring lines. 2. The isozyme patterns of COD 12days'seedling leaves can be used to mark the four male-sterile lines and the corresponding maintainer line. The isozyme patterns of COD 9days'seedling leaves can be used to identify the male-sterile lines and the maintainer line. 3. The SDS-PAGE electrophoretogram can be used as the biochemical markers of male-sterile lines and the corresponding maintainer line. 4. The male sterile had been decided in very early in the seedling development. The 9day is the key stage of the sterility gene expression in CMS in wheat. 5. It had be found that development of pollen about ms(Ae.ventricoca)-90-110 is very different of the other male sterile lines by examination microscopic and it'abortion is earlier and severer. This phenomenon can be verified each other by the isozyme patterns of COD. 6. The abnormal expression of COD is one of the cause of the pollen abortion. 7. The 75kD polypeptide in SDS-PAGE about all protein in eight isonuclear alloplasmic and allonuclear isonuclear male-sterile lines had been found keeping close with the male sterile.
