Peach (Prunus persica L.) is one of the most extensively cultivated temperate zone fruit tree in the world. Peach is original from China, which genetic resources are rich, and it has long cultivated history. The peach variety is abundant, and has extensive adaptability to the ambient conditions. Peach is an important unwoody tree. Peach is cultivateed extensively in our country. Peach is one of the nutrition-healthy fruit received by many people, and it tastes fragrant and sweet, and its nutrition is abundant, which contained many kinds of amino acid human body can not synthetis. Peach crust can be done as the active carbon, and peach kernel can squeeze industry oil, and peach still can be used as medicine and many kinds of commodity. But peach is fairly sensitive to the environment, and region distribution of it is obvious.The autumn of the fresh peach is short and concentrated, and its pulp is easily soft .The utmost point of peach is unable to endure the storage and transportation. Peach is difficult for sale abroad in ripe period, and the economic value is not high after it is softened. Gansu Province is its place of origin and one of the fittest cultivated area. In order to solve the problem in production of the seedling and promote the development of peach industry, the experiment was designed to study on high efficient micro propagation of different-maturing peach varieties and offered the necessary technology in developing virus-free peach seedling. The result showed as follows: 1. Experiment of the bud induction of fresh meal peach varieties was conducted by using the orthogonal experiment design. The results showed that the suitable medium for the bud induction from shoot tips was MS+1.0mg/L6-BA+ 0.5mg / LGA3+2.0mg / LIBA. 2. Dark treatment and low temperature were needed in the initiation phase stimulated rooting.1/2MS cooperated with soak of 100mg/LIBA and 16℃dark treatment in 8 days was the best to induce rooting. There was no relationship between soaking time and rooting rate. We also found that 22℃was suitable for the rooting of test-tube plantlet. 3. Through reducing humidity gradually and three-step acclimatization procedure, the survival rate of plantlets with well-developed shoots and roots was over 80%. 4. Result of photosynthesis indicated that the photosynthesis rate of rooting plantlet was clearly higher than that of the test-tube plantlet without rooting (chongyanghong unclear), and the photosynthesis rate of the plantlet without rooting rise and fall from top to bottom at 0 umol / m2s. 5. The protective cell chloroplasts and SDS polyacrylamide gel electrophoresis were employed to analyze the genetic stabilization of four peach varieties. The result showed that four peach varieties'band had no difference with the band of four peach varieties cultured 8th. It showed that tissue culture can keep their specific genetic stabilization, and high efficient micro propagation system had high genetic stabilization by way of "adventitious bud type". 6.The result also showed that the average chlorophyll content of peach test-tube plantlet was 19.32mg / L, its water potential was( ? w ) -0.403Pa, and transpiration rate was 3.25 mg/㎡s, equivalent to 63.68%, 64.20% and 24.04% of maternal plant respectively.
