| As a region different from other epidemic regions, the Chinese region is the biggest and unique epidemic region of Puccinia striformis f.sp. tritici in the world. Presently, the better methods against the wheat stripe rust are using resistance cultivars and chemicals. But whatever measures will be taken, the first step is to observe the virability of virulence and the structure of the populations of Puccinia striformis f.sp. tritici in shortest time. From the view of the virulence, the study was be taken to find DNA polymorphisms in 17 races with amplified fragment length polymorphism(AFLP) and simple sequence repeat (SSR or microsatellite markers). The results as follows:1. A 285bp polymorphism band of CY24 was been obtained near the 278~281bp amplified region, when the SSR primer RJ17 was used to analyze the experimental 12 isolates. Then the band was affirmed with 6 added isolates.2. A 182bp polymorphism band of CY31 was been obtained near the 170~176bp amplified region, when the SSR primer RJ21 was used to analyze the 12 experimental isolates. Then the band was affirmed with 6 extend isolates.3. A 650bp polymorphism band of CY29 was been obtained, when the 30 AFLP primer combinations were used to analyze the 30 experimental isolates. DNA polymorphism was obtained in 12 of 30 combinations.4. A 310bp polymorphism band of correlative Yr9 virulence was been obtained, when the SSR primer RJ20 was used to analyze the 11 experimental isolates. Then the band was affirmed with 7 extend isolates.5. A 308bp polymorphism band of correlative SU1 lseries virulence was been obtained, when the SSR primer RJ3 was used to analyze the 11 experimental isolates. Then the band was affirmed with 5 extend isolates. |
