Effects of several factors, such as media, types of explants, and plant growth regulators on in vitro culture and rapid propagation of Cornus alba 'Aurea' were investigated in detail in the thesis. The axilla-buds from the cuttings of two-year-old branchs picked in June were the optimal explants. The optimal initial medium is MS basal medium supplemented with 6-BA (0.50 mg/L) and IBA (0.05-0.20 mg/L) and the rate of sprouting was up to 87.4% accordingly. In the initial course of the explants, the serious browning occured. It was discovered that the site of the bud explants was the main influential factor of browning. The explants differentitated a lager number of young buds on M basal medium containing 6-BA (0.50 mg/L) and IBA(0.05 mg/L). On the above medium shoots multiplicated by 4.74-fold after 20 days of culture. After analizing the influential fators of culture, the optimal combinations for reserving and preventing vitrification in in vitro regeneration of Cornus alba 'Aurea' were abtained. The regenerated shoots were rooted on the 1/2 strength M basal medium supplemented with 0.50 mg/L IBA and the rate of rooting could reached to 91.7%. The complete plantlets were transplanted into substrate (perlite:vermiculite 1:1) with the survival rate up to78.9% .Rooting was maximized (91.7%) for handwood cuttings of Cornus alba 'Aurea' treated with 100mg/L ABT for 5 minutes. An ideal medium for rooting cuttings was sand and the temperature at 23-28 C. |
