Characterization Of New Wheat Lines Highly Resistant To Stripe Rust Drieved From Hexaploid Triticale×Wheat

Abstract/Summary:
Rye (Secale cereale L.) offers many useful genes for common wheat (Triticum aestivum L.) improvement. Hexaploid triticale possesses all R genome chromosomes of Secale, which can be used as an important 'bridge' for transferring rye genes into wheat. In the hybrids of triticale X wheat, all R and D-genome chromosomes were present as univalents and were given a chance to misdivide, resulting in a very high frenqency effusion. By this way, translocations, substitutions and additions with rye genes can be easily produced in wheat genetic background. Moreover, desire genes in A, B-genome chromosomes of triticale can be also transeferred into common wheat by recombination of homologous chromosomes. From the hybrid progenies of triticale wheat, new wheat lines NR98116-8-18, NR98116-9-2 and NR98116-9-3 with high resistance to stripe rust were obtained. In present studies, we characterized these new wheat lines with high stripe rust-resistance on Karyotype, chromosome pairing of PMC MI, Giemsa-C banding, Genome in situ hybyridization and SDS-PAGE. The results were as follows:1. NR98116-8-18, NR98116-9-2 and NR98116-9-3 derived from the hybrid of combination of Currency / chuanyu 12 // xiaoyan 6 /// A302 showed high resistance to stripe rust. Common wheat xiaoyan 6, chuanyu 12 and A302 exhibited high or middle susceptivity to stripe rust respectively, while triticale Currency exhibited high resistance. These suggested that the high stripe rust-resistance in Currency had been transferred into NR98116-8-18, NR98116-9-2 and NR98116-9-3.2. By analysis of Karyotype, chromosome pairing of PMC MI, Giemsa-C banding and Genome in situ hybyridization, it was showed that NR98116-8-18 was wheat-rye translocation lines, NR98116-9-2 was 3R/3D disomic substitution lines and NR98116-9-3 was 3R disomic addition lines.3. The result of SDS-PAGE revealed that NR98116-9-2 and NR98116-9-3 possessed six and seven HMW-GSs respectively, of which HMW-GS 6.8 encoded by Glu-Bl was derived fromCurrency. Wheat with seven HMW-GSs was unexpected, which was not reported previously. It was also suprising that the protein contents of NR98116-9-2 and NR98116-9-3 reached up to 20.08% and 16.88%, respectively, higher 69.2% and 42.2% than local check Chuanmai 107.
